Objectives of the study


Generally, this study aims to observe the process of

endocytosis in single-celled bacteria. Specifically, this study aims to:


Observe the phagocytosis of Tetrahymena sp. in graphite

shavings Observe the phagocytosis of Tetrahymena sp. in India ink Compare the number of food vacuoles and the manner of feeding of Tetrahymena sp. in two different food source

Materials and Methods


Materials
Tetrahymena sp. cell Microcentrifuge tubes, 1.5ml Micropipettors and tips Graphite shavins from

suspension Chlorella sp. Culture Microscope Slides and Cover Slips Formalin Solution, 3% Filter paper strips

pencil India ink, 1% Toothpicks Pencil Razor blade/cutter

Materials and Methods


Methodology On Graphite Shavings -A drop of Tetrahymena sp. was suspended in a slide. Shavings of graphite from a lead pencil were introduced to the bacteria on the slide. A cover slip was placed on the Tetrhymenna sp. culture and graphite. The phagocytocis of the Tetrahymena sp. was observed under the microscope on an HPO.

Materials and Methods


On India ink -An equal amount of the Tetrahymena sp. suspension and 1% India ink was mixed in a microcentrifuge tube. The a small amount of the mixture was pipetted on the slide and was observed under the HPO objective of the microscope. Comparison -The manner of Tetrahymenna sp.s feeding and swimming on a graphite shavings and an India ink solution was compared. Food vacuoles of the cells were observed again, under the HPO of the microscope. Number, location, and appearance of food vacuoles and cilia were observed and noted.
