PHARMACEUTICAL ENGINEERING APPLICATIONS B TUTORIAL #5/2013 5.1 The enzyme, beef catalase has been used to accelerate the decomposition of hydrogen peroxide: 2H2O2 + beef catalase 2H2O + O2 + beef catalase (S) + (E) The following table gives data on the concentration of H2O2 as a function of time for a reaction mixture with pH = 6.76 and constant temperature = 30C in a batch reactor. t (min) [S] (mol dm-3) 0 10 20 0.0158 50 0.0106 100 0.0050 0.0200 0.01775

(a) Determine the Michaelis-Menten parameters, Vmax and KM. (b) If the total enzyme concentration is increased threefold, determine what the H2O2 concentration would be after 20 minutes. 5.2 Saccharomyces cerevisiae or bakers yeast (the common name for the strains of yeast commonly used as a leavening agent in baking bread) contains enzymes that cause the breakdown of the starch in the bread dough. Data on the rate of uptake of oxygen (substrate, S) by bakers yeast (enzymes, E) during aerobic growth in a particular medium at 23.4C, without and with the addition of a fixed amount of sulphanilamide (inhibitor, I) are shown in the table below. Partial pressure, PO2 (mm Hg) 0.0 0.5 1.0 1.5 2.5 3.5 5.0 -rO2 (without I) (mL O2/hr/mg of cells) 0.0 23.5 33.0 37.5 42.0 43.0 43.0 -r O2 (with I) (mL O2/hr/mg of cells) 0.0 17.4 25.6 30.8 36.4 39.6 40.0

(a) Determine the Michaelis-Menten parameters, Vmax and KM. (b) Use the Lineweaver-Burk plot to determine the type of inhibition caused by sulphanilamide.
