1. PURPOSE: To find patients Rhesus blood group.

2. RESPONSIBILITY : Medical Laboratory Scientist/Medical Laboratory Technician.

3. REAGENTS/EQUIPMENT :

Khan tubes
Pasteur pipettes
Centrifuge
Blood grouping racks
Saline wash bottles

4. PRINCIPLE

Based on the principle of agglutination of D-antigen positive red cells in the presence of
antibody Anti D directed towards the antigen.

5. PROCEDURE

Label tube with a laboratory number and test identification (Anti D).
Place two drops of anti D in a tube labeled anti D.
Add to the tube red cells to be tested.
Centrifuge at 1500rpm for 30 seconds and observe for agglutination.

6. RESULTS:

(a) Agglutination of red cells Rh positive
(b) No red cell agglutination Rh negative

If the result is Rhesus negative perform D

Incubate negative Rhesus test at 370C for 15 minutes and perform the Indirect
Antiglobulin Test (see IAT SOP).
If the results are negative then the patient is truly Rh(D) Negative.

positive patients are given Rh D negative blood if transfusion is
required.
