1. PURPOSE: To find patients blood group.

2. RESPONSIBILITY : Medical Laboratory Scientist/Medical Laboratory Technician.

SPECIMEN : EDTA or clotted blood sample can be used.

3. EQUIPMENT :
75mm glass or disposable test tubes.
Pasteur pipettes
Blood grouping racks
Centrifuge
Saline wash bottles

4. PRINCIPLE
Based on the principle of agglutination. Red cells with the antigen will agglutinate when
tested against the corresponding antibody.

5. METHOD

Cell Grouping
1. Label tubes with laboratory number and test identification (anti A, B and AB).
2. Prepare cell suspension for cells being tested.
3. Place two drops of anti A, anti B, and anti AB reagent in the appropriately labeled
tubes.
4. Add to each tube one drop of 2-5% cell suspension (in normal saline, serum or
plasma of the red cells to be tested).
5. Centrifuge at 1500rpm for 30seconds and observed for agglutination.

Serum Grouping
1. Label the tubes with test identification (A, B and O).
2. Add two drops of test serum in all tubes in the corresponding column.
3. Add one drop of 2-5% group A cell suspension in tube labeled A.
4. Add one drop of 2-5% group B cell suspension in tube labeled B.
5. Add one drop of 2-5% group O cell suspension in tube labeled O.
6. Centrifuge at 1500rpm for 30seconds and observe for agglutination.
