IDENTIFICATION OF BIOACTIVE FLAVONOID COMPOUNDS

BASED ON GC-MS DATA IN PROPOLIS

ABSTRACT
Background :
Flavonoids are the major bioactive constituents in Propolis and have the potency as a anti-
inflammatory, antioxidants, and antimicrobial agents. To optimize potency of Propolis, it is
required the identification of the flavonoids as markers of major bioactive compounds
contained in Propolis.
Objective :
The purpose of this research is to identify of flavonoids contained in Propolis using
spectrophotometry and GC-MS methods.
Method :
This research used GCMS method to analyse the flavonoids. The use of GC-MS method was
successful to identify the flavonoid constituents qualitatively, as compared to the Wiley NIST
data library. The content of total flavonoids was evaluated using uv spectrophotemetry, based
on an established procedure.
Result
GC-MS analysis revealed that propolis contains 43 unknown compounds. From the 43
compounds, there are several compounds having high level similarities with known
flavonoids and non flavonoid compounds, namely, pinocembrin (quality = 95), 2-propanol
(quality = 90), guaiol (quality = 98), 2-napthalenemethanol (quality = 98), cadinene (quality
= 97), chrysin (quality = 94), and azacyclotridecan-2-one (quality = 92). Analysis of total
flavonoids based on uv spectrophotopmetry method was 0.343 % (b/b).
Conclusion
The main active compounds of Propolis are flavonoids. The flavonoid was identified
pinocembrin based on GCMS data as compared to the available data base. The total
flavonoid content was 0.343% based on spectrophotometric analysis. High flavonoids
compounds in propolis makes it a promising agent as antiimflamation, antioxidant and
antimicrobe.
