DEXTRAN MEASUREMENT

Spectrophotometric Method

Take 60 ml Juice in a 100 ml beaker.

Add 1~2 grams of LIWATIT MP-7080 (Anion) + 1~2 grams
LIWATIT CP- 3050 (Cat ion)
Shake with magnetic stirrer for 10 minutes.
Filter the juice through cloth or 100 Mesh wire netting.
Add 10 ml Trichloro Acid and mix.
Filter the solution through Whattman No.1 Filter Paper
Take 12.5 ml Filtrate and add 12.5 ml ethanol drop wise by mixing.
Fill the solution in cell tube for Spectrophotometric reading at 720
nm.
Read the absobence reading at 720 nm.
Look up the mg dextran in the curve chart against absorbancy reading.

Calculation = mg. Dextran x 1000 x 100 = Dextran PPM

12.5 x brix of juice
Example:
Juice Brix = 15.50
Absorbancy in 10mm(1cm) Cell = 0.0588
Now according to the chart
0.0588 Absorbancy in mg Dextran is = 5.60
Now Calculate in PPM.
5.60 x 1000 x 100 = 2890 PPM Dextran
12.5 x 15.5
