A Few Things about Protein Engineering

Protein engineering is the method of developing helpful proteins. This discipline


emphasize on analysis of the understanding of protein folding and protein design
principles. The core programmed of this discipline is the use of recombinant DNA
technology to change amino acid sequences. The study was first emerged in the
early 1980. The main subject areas include DNA synthesis, X-ray crystallography,
gene synthesis, enzyme, computer modeling of protein structure and protein
folding.
The basic assumptions of protein engineering are:
a. Protein is not at an optimum for the function. Design of protein much
depends on the interaction of molecules within the surrounding.
b. The accommodation of the required sequence is possible without folding or
disrupting the structure of protein; and
c. There is not much significant difference between the new sequence and the
native sequence.
The discipline adopts two major approaches, i.e. rational design and directed
evolution. While the former deals with the knowledge and structure of protein; the
later random mutagenesis is applied to a protein to get desired qualities. The
desired changes are caused by mutations and this does not required any predetermined structure of the protein.
