RESULT

There is no result from this 4B experiment (MTT Assay) because bead


prepared from last experiment was contaminated.

DISCUSSION
Contamination of cell cultures is the most common problem encountered
in cell culture laboratories, sometimes with very serious consequences. Cell
culture contaminants can be divided into two main categories, chemical
contaminants such as impurities in media, sera, and water, endotoxins,
plasticizers, and detergents, and biological contaminants such as bacteria,
molds, yeasts, viruses, mycoplasma, as well as cross contamination by other cell
lines. While it is impossible to eliminate contamination entirely, it is possible to
reduce its frequency and seriousness by gaining a thorough understanding of
their sources and by following good aseptic technique
Bead contamination can occur when the process of preparing the bead not
aseptically done. Bead can contaminated during the process of mixing alginate
and media, if the mixture of alginate and media leave to long without the cell,
there might be loss of nutrient. The wrong technique of using pipette can also
cause damage to the bead and cell because cell might damage. The
contamination also can occur during the changing of media, during changing the
media the bead are exposed to unsterile environment.
