Spectra Optia System Common Operations

1.0 PRINCIPLE/PURPOSE: To describe the procedure to perform common operations of the
Spectra Optia cell separator. The Spectra Optia system separates and collects blood
components from donors and patients for cell therapy, therapeutic apheresis, and transfusion
medication applications. Refer to the SOP for the specific procedure performed for more
detailed instructions.
2.0 SCOPE: This procedure will be performed by trained Apheresis staff or trained designee.
3.0 MATERIALS:
3.1 Spectra Optia disposable tubing set( Spectra Optia Collection Set for Stem Cell
Collection, Spectra Optia Exchange Set for Red Blood Cell Exchange)
3.2 Fluid warming set
3.3 Anticoagulant Citrate Dextrose Solution A (ACDA)
3.4 1000ml bag of NaCl 0.9% (Normal Saline)
3.5 Alcohol pads
4.0 EQUIPMENT:
4.1 Spectra Optia Cell Separator
4.2 HOTLINE blood warmer
5.0 QUALITY CONTROL:
5.1 Perform an alarm test then prime the disposable tubing set and before hooking up
anyone to the system.
5.2 Maintenance must be performed on the Spectra Optia cell separator. See SOP
APH:C003
6.0 PROCEDURE:
6.1 Setting up the cell separator
6.1.1 Turn the power switch to the on position.
6.1.2 Touch Select procedure. The procedure selection screen appears.
6.1.3 Select the type of procedure that will be performed, then touch confirm.
The system loads the procedure software.
6.2 Unpacking the Lines and the bags
6.2.1 Use Collection Set for Mononuclear Cell Collection and use Exchange
Set
for Red Blood Cell Exchange.
6.2.2 Verify the tubing set has not expired by checking the expiration date on
the
cover of the package. Record expiration date and lot number on the
procedure record.
6.2.3 Put the tubing set package on top of the centrifuge cover with the
package
label upright and facing the operator, and remove the cover from the
package.
6.2.4 For Exchange Set:
6.2.4.1 Take out the spike holder. The spike holder holds the replace
line (white spikes), the AC line (orange spike), the saline line
(green spike), and the extra remove line (yellow clamp). The
remove bag with attached remove line is located inside the
spike holder.

6.2.4.2 Take out the remove bag and the extra remove line, and put them
on the right side of the centrifuge cover.
6.2.4.3 Take out the vent bag and hang the bag on the left side of the IV
pole.
6.2.4.4 Remove the replace line and clamp one of the lines to the spikes.
Put the line on top of the remove bag.
6.2.4.5 Take out the AC line (orange spike) and the saline line (green
spike) and hang the lines over the left side of the system.
6.2.4.6 Take out the coiled inlet line (red clamps) and remove the paper
tape from the coil. Hang the inlet connection on the left end of the
IV pole. Repeat this step with the return line (blue clamps).
6.2.4.7 Take out the cassette and the channel, and put them on top of the
centrifuge cover.
6.2.4.8 Discard the empty tubing set package in recycle bin.
6.2.5 For Collection Set:
6.2.5.1 Take out the product bags and the vent bag and put them on the
right side of the centrifuge cover.
6.2.5.2 Take out the coiled inlet line (red clamps) and remove the paper
tape from the coil. Hang the inlet connection on the left end of the
IV pole. Repeat this step with the return line (blue clamps).
6.2.5.3 Take out the AC line (orange spike) and the saline line (green
spike) and hang the lines over the left side of the system.
6.2.5.4 Take out the cassette and the channel, and put them on top of the
centrifuge cover. Discard the empty tubing set package in recycle
bin.

6.3 Loading the Cassette into the Cassette Tray
6.3.1 Put the bottom of the cassette in the bottom edge of the tray.
6.3.2 Ensure that there is nothing lodged behind the cassette or the tray that could
interfere with the loading.
6.3.3 Press the top corners of the cassette to snap the cassette into the tray.
6.3.4 Hang the bags on the IV pole. If using an Exchange Set, hang the remove
bag on the IV pole. If using a Collection Set, hang the bags from left to right
on the IV pole in this order: Vent bag, Plasma bag, Collection bag.
6.4 Loading the Channel in the Centrifuge
6.4.1 Open the Centrifuge door.
6.4.2 Locate the pin on the filler latch. Raise the latch by pushing the pin toward
the center of the filler while pulling up the latch.
6.4.3 Turn the centrifuge so that the loading port faces the operator.
6.4.4 Fold the channel in half. If using a Collection set, tuck the chamber inside
the folded channel.
6.4.5 Push the channel up through the loading port and the opening in the center
of the filler. Lower the filler latch and lock it in place.
6.4.6 Position the lower collar in the filler latch so that the pink line is not
obstructed by the other lines.
6.4.7 Push the collar into the collar holder until you hear the click of the locking
pin as it pops out and locks the collar in place. Ensure that the notch at the
base of the locking pin is visible. If the collar is locked, you can see the
notch.
6.4.8 Starting with the connector, insert the channel into the groove in the filler,
finishing with the inlet port. Run your finger over the groove and push down
any section of the channel that is not completely inserted in the groove. The
channel must sit flush with the groove.
6.4.9 If using a Collection set, put the chamber into the chamber bracket. Ensure
that the chamber sits behind the clip at the opening of the bracket, so that
the clip retains the chamber in the bracket. Put the collect line to the

chamber behind the lip of the optical reference, so that the line does not
block the optical reference.
6.5 Loading the Lower and Upper Bearings, and the Upper Collar
6.5.1 Insert the narrow part of the lower bearing into the lower bearing holder, and
the narrow part of the upper bearing into the upper bearing holder. Ensure
that the braided section of the loop is not twisted.
6.5.2 Position the upper collar below the upper collar holder and insert the line into
the holder. Pull the line up to secure the upper collar in the holder.
6.5.3 Spin the centrifuge clockwise to ensure that it rotates freely.
6.5.4 Close the centrifuge door.
6.5.5 Touch load. The system lowers the cassette.
6.6 Testing the tubing Set
6.6.1 If using an Exchange Set, follow the instructions on the screen to perform
the following steps: Close the inlet saline line and the return saline line.
Clamp the extra remove line. Touch continue.
6.6.2 If using a Collection Set, follow the instructions on the screen to perform the
following steps: Clamp the line to the diversion bag. Close the inlet saline
line and the return saline line. Clamp the line to the sample bulbs. Touch
continue.
6.6.3 Follow the instructions on the screens to clamp the inlet line and the return
line.
6.6.4 Touch Continue. The system performs several tests to verify the the tubing
set and system components function properly, and the tubing set is loaded
correctly.
6.7 HOTLINE Set up
6.7.1 Connect the male luer connection on the return line of the Spectra Optia
system disposable tubing set to the female luer connection on the fluid
warming set.
6.7.2 Secure but do not over tighten the connections.
6.8 Priming the Tubing Set
6.8.1 Follow the instructions on the screen to perform the following steps: Spike
the fluid containers, fill the drip chambers, insert the AC line in the AC fluid
detector..
6.8.2 Touch Start Prime. The system primes the AC line.
6.8.3 Follow the instructions on the screen to open the inlet saline line and the
return saline line.
6.8.4 Touch Continue. The system primes the return line, the inlet line, the RBC
line, and the channel. The system primes the set in about 3 to 4 minutes.
When the system finishes the prime it sounds a tone. The patient data
screen appears. The menu buttons at the top of the screen are active so that
you may enter or change patient or procedure data during the prime.
6.8.5 Follow the instructions on the screen to continue the procedure.
6.9 Rinseback
6.9.1 To finish the procedure, Touch the Rinseback tab, then touch Proceed to
Rinseback, then touch Confirm. The screen will prompt you to clamp the
inlet and open the inlet saline line.
6.9.2 For Red Blood Cell Exchange the run target screen appears when a run
target is attained. A tone sounds, the pumps pause, and the centrifuge
continues to spin until Disconnect is touched. Rinseback is not
recommended.
6.10 Removing the Tubing Set
6.10.1 Follow the instructions on the screen to disconnect the patient and unload
the tubing set.
6.10.2 Open the centrifuge door.
6.10.3 Remove the upper collar from the collar holder by grasping the lines above
and below the collar, and pulling the lines downward.
6.10.4 Remove the upper and lower bearings from the bearing holders.

6.10.5 If you are using a Collection Set, remove the chamber from the bracket.
6.10.6 Gently pull the channel from the filler.
6.10.7 Push in the locking pin on the centrifuge collar holder and remove the collar
from the holder by grasping the tubes above the collar and pulling upward.
6.10.8 Push the filler latch pin toward the center of the centrifuge, and raise the
filler latch.
6.10.9 Fold the channel in half and pull the channel through the loading port and
out of the centrifuge chamber.
6.10.10 Lower the filler latch.
6.10.11 Close the centrifuge door.
6.10.12 Remove the lines from the fluid detectors.
6.10.13 Remove any bags from the IV pole.
6.10.14 Press the latch on the upper right corner of the cassette tray, and lift the
cassette from the tray.
6.10.15 Discard the tubing set in the biohazard bin.
7.0 PROCEDURAL NOTES:
7.1 Before each use of the Spectra Optia system, inspect all tubing especially in the
centrifuge and on the front panel to ensure that it is not kinked. Tubing that is
occluded or partially occluded, can lead to malfunction or fluid imbalance.
