This assignment is due the week of 9/15 9/18 at the beginning of your lab section. It should
be typed using Arial or Times New Roman font (11 of 12pt). There are three parts to the
assignment, include a heading for each part. All figures should include a figure number and
legend (descriptive title and brief caption) below the graph. Graphs should be generated using
Excel, and properly scaled with fully labeled axes, including units where applicable.
Part I: Introduction to Spectrophotometry

(6 pts)

1. Figure 1. Absorption Spectra: Plot the 4 sets of data from Table 6-2 (p. 47) on a single
graph. Include a key identifying the 4 data sets. Identify and mark the lambda max for
each colored sample. (4 pts)
2. Define lambda max and discuss the importance of its use generating standard
curves. (2 pts)
Part II: Paper Chromatography Analysis and Identification of Amino Acids

(6 pts)

1. Generate a table that includes the information listed in Table 8-1 (p. 72). Include all 6
amino acids listed and the unknown you tested (either X or Y). Identify which amino
acids were present in your assigned unknown. (3 pts)
2. Conclusion: Write a one paragraph conclusion regarding this experiment. Discuss the
findings that support the identification of the amino acids in your assigned
unknown. (3 pts)
Part III: Quantitative Analysis of Protein The Biuret Test

(8 pts)

1. Turn in pages 74 and 75 from your lab manual (1 pt)


2. Figures: Based on the data from Tables 8-2 and 8-3, generate two graphs:
a. Figure 2. Absorption Spectra: Based on the data from Table 8-3, generate a
graph and indicate where lambda max is located on the plot. (2 pts)
b. Figure 3. Standard Curve: Based on the data in Table 8-2, generate a standard
curve for BSA. Make a scatter XY plot; add a trendline and the equation for the
trendline, and the R-squared value. (2 pts)
3. Results: Include the heading Results, compose a concise paragraph summarizing what
you determined from this investigation. State which unknown (A, B, C or D) you tested,
how much BSA the unknown contained, and how you determined this value. Refer the
reader to each of the figures outline above in this text. (3 pts)
