CellwalldeficientbacteriaReferredtoasLformshavelosttheabilitytoyourtomaintainorbuildarigid
peptidoglycanenvelope.Wehavegeneratedstable,nonrevertingLformvariantsoftheGram
positivepathogenListeriamonocytogenes,andStudiedthecellularandmolecularchangesassociated
withthistransition.StableLformcellscanoccurassmallprotoplastsarelikevesiclesandas
multinucleated,largebodies.Theyhavelostthethick,multilayeredmureinsacculusandare
surroundedbyacytoplasmicmembraneonly,althoughpeptidoglycanprecursorsarestillproduced.
WhiletheylackmureinassociatedmoleculesIncludingInternalAmembraneanchoredproteins
suchasinternalBareofretained.Surprisingly,Lformswerefoundtobeabletodivideandpropagate
Indefinitelywithoutawall.TimelapsemicroscopyoffluorescentlylabeledLformsIndicateda
switchtoanovelformofcelldivision,thewheregenomecontainingmembranevesiclesarefirstFormed
withinenlargedLforms,andsubsequentlyreleasedbycollapseofthemothercell.Arraybased
transcriptomicsofparentandLformcellsRevealedmanifolddifferencesinexpressionofgenes
associatedwithmorphologicalandphysiologicalfunctions.TheLformsfeaturedownregulated
metabolicfunctionscorrelatingwiththedramaticshiftinthesurfacetovolumeratio,Whereas
upregulationofstressgenesreflectstheDifficultiesinadaptingtothisunusual,cellwalldeficient
lifestyle.
