Learning Outcomes:

1. Define and Give examples of Restriction Endonucleases.

2. Explain how restriction enzymes produce Recombinant DNA.

Enzymes called restriction endonucleases

revolutionised molecular biology because
of their ability to cut DNA at specific
sites.
But How DO restriction enzymes work?

They allow the creation of recombinant molecules; these

enzymes recognise a specific DNA sequence, ranging from
and cut the DNA at a specific base within this sequence.

The Recognition sites for many Restriction Enzymes are

palindromes...
A palindrome is a word or phrase
that reads the same forward and
in reverse, e.g...

SO!... The palindromic DNA sequence reads the same

from 5' to 3' on one strand
as it does on the complementary strand.

Cutting the DNA at the same base on either strand

produces staggered cuts Called sticky ends.

These short, unpaired

sequences are the same for
any DNA that is cut by this
enzyme...
SO, These sticky ends allow DNA from different sources
to be easily joined together!

Describe this Diagram!! Work in pairs, You have 5 minutes to provide a

step by step, explanation of what it is going on! (use the animation to help!)
