Lab title:Growth conditions Physical O2 and Temperature

Objective: The objective of this experiment is to observe the growth of P.aueroginosa, S.aures,
C.sporanges, and E.coli in thioglycollate broth tubes, and in aerobic and anaerobic environments
using nutrient agar plates. The second objective of this experiment is to place S.marcescens,
S.epidermidas, G.stearothermophilus and S.cerevisiae on nutrient agar plates and SDA plates and
are incubated in various temperatures to observe the growth of the bacteria.

1. Our results:
Ecoli: aerobe/ anaerobe
P.aeruginosa: aerobe/anaerobe
S.aureus: aerobe/anaerobe
C.sporanges: anaerobe

2. Yes, our results agree with researched results because E.coli, P.aeruginosa, and S.aureus
are facultative anaerobes which means they use aerobic respiration to produce ATP if
oxygen is present but is capable of anaerobic respiration if oxygen is absent. C.sporanges
was expected to be anaerobic, which means it is capable of producing ATP when oxygen
is absent.

1. Results:
Serratia marcescens: Mesophile
Staphylococcus epidermidis: Mesophile
Bacillus stearothermophilus: Thermophile
Saccharomyces cerevisiae: No growth
2. The results for Serratia marcescens and Staphylococcus epidermidis were correct as they
were expected to be mesophiles. Serratia marcescens grows best around 20-45C.
Staphylococcus epidermidis has optimal growth temp of 26-37C. Since mesophiles
typically grow in moderate temperatures of 20-45C they classified as such.
The results for Bacillus stearothermophilus was correct as it has a typical growth range
from 30-75C and was expected to be a thermophile. Since thermophiles in relatively hot
conditions rougly between 41-122C it classified as such.
The results for Saccharoymyces cerevisiae were incorrect as this was expected to be a
mesophile but there was no growth.

Lab 7 questions:
1. Resazurin is added as an oxidation-reduction indication. In an oxidized state it turns pink,
and in reduced state it is clear. Agar is added to help with initiation and growth of
anaerobes that are small by impending diffusion of oxygen into medium.
2. The red layer indicated oxidation of the resazurin. To remove this red colour you heat the
medium.
3. Thermophiles typically grow in relatively hot conditions such as a hot spring. Examples
of these bacteria are Geobacillus stearothermophilus and Thermococcus litoralis.
Psychrophiles typically grow in relatively cold environments such as deep sea,
mountains, and Polar Regions. Examples of these bacteria are Chryseobacterium
greenlandensis and Pseudomonas fluorescens.
