Problems Lecture 13

1. Assign a function or product for each of the following structures or pathways

a. chlorophyll absorbs light energy
b. Photosystem I uses light energy to reduce NADP+ to NADPH
c. Photosystem II uses light energy to oxidize water and create a proton gradient
d. Plastoquinone delivers electrons from PSII to cytochrome bf
e. Plastocyanin delivers electrons from cytochrome bf to PSI

2. The three electron transporting complexes of the thylakoid membrane can be called
plastocyanin-ferredoxin oxidoreductase, plastoquinone-plastocyanin
oxidoreductase, and water -plastoquinone oxidoreductase. What are the more
common names for these enzyme complexes?

plastocyanin-ferredoxin oxidoreductase = PSI
plastoquinone-plastocyanin oxidoreductase = cytochrome bf
water -plastoquinone oxidoreductase = PSII

3. When the antifungal compound myxothiazol is added to a suspension of
chloroplasts, the PQH2/PQH ratio increases. Where does myxothiazol inhibit
electron transfer?

The increase in PQH2 happens because transfer of electrons to cytochrome bf is
blocked.

4. Plastoquinone can freely diffuse laterally throughout the thylakoid membrane in
keeping with its function as an electron carrier. What does this tell you about its
structure?

It must have a hydrophobic portion that is able to diffuse through the lipid bilayer.
(Plastoquinone has a hydrocarbon side chain of various lengths up to about 45
carbons.)
