STUDY THE EFFECT OF CONVULSANTS AND ANTICONVULSANTS IN MICE OR RATS.

Chemicals Pentylene tetrazole (Leptazole). Dose-80 mg per kg subcuteneously. Trimethadione-400 mg per kg Diazepam-4 mg per kg intraperitoneally

Theory Convulsion can be produced in mice by injecting analeptics like strychnine (1 mg per kg) or pentylenetetrazol (Leptazol). Drugs that are effective in electrically induced seizures are found to be beneficial in grand-mal epilepsy where as those effective in chemically induced seizurcs are found to be useful in petit-mal epilepsy.Anticonvulsants are antiepileptic agents. Antiepileptic drugs cause normalization of seizure foci, prevention of post-tetanic potential, potentiation of preor postsynaptic inhibition and prolongation of the refractory period.Trimethodione blockschemically induced seizures and is used in treatment of petitmal epilepsy. It increases threshold of excitability in thalamus and depresses polysynaptic transmission. Dizepam also prevents chemically induced seizures and suppress spread of seizures. It stimulates the GABA receptors. Procedure Select six healthy rats or mice. Two of them are fed trimethadione (400 mg per kg) orally. After one hour both mice/rats are injected pentylene tetrazole intraperitoneally. Another two mice/rats are injected diazepam (4 mg per kg)intraperitoneally. After half an hour all the rest four mice/rats are injected pentylene tetrazole intraperitoneally.Record the presence or absence of convulsions. Abolition of both tonic and clonic seizures in 50% of animals is criteria for anticonvulsant activity of drugs.
