FILAMENTOUS HEMAGGLUTININ
Filam entous hem agglutinin (fim brial hemagglutinin or FHA) is one of two hem agglutinins produced by phase I strains of Bordetella pertussis. 1 The use of FHA as one com ponent of a new acellular vaccine is currently under investigation. FHA is nam ed for its rod-like shape and its ability to agglutinate erythrocytes from a variety of species.2 On a weight basis, FHA is five to seven tim es m ore active in hem agglutination (HA) assays than is pertussis toxin.1,3 Moreover, the HA activity of FHA, but not that of pertussis toxin, is abolished in the presence of low levels of cholesterol.2,3 FHA is a protein with an approxim ate m olecular weight of 200,000 daltons. However, som e preparations of this protein contain varying am ounts of two lower m olecular weight com ponents in the 90,000 to 130,000 dalton range. Although m onoclonal and polyclonal antibodies prepared against FHA also react with these faster m igrating com ponents, it is not known whether the latter represent degradation products or subunits of FHA. 4 This product is intended for research purposes only and is not intended for use in humans. W hen run on SDS-polyacrylam ide gels according to W yckoff,5 a m odification of the Laem m li6 gel system , FHA from List Biological Laboratories exhibits a m ajor band and several m inor bands of lower m olecular weight, believed to be related to the intact m olecule. Each lot is tested for its ability to agglutinate goose red blood cells. This HA activity is inhibited by either anti-FHA or cholesterol. W hen exam ined in W estern blotting experim ents, FHA from List Biological Laboratories reacts strongly with anti-FHA hybridom a supernatants. FHA is aseptically packaged and is provided as a lyophilized powder. W hen reconstituted with 0.5 m l of sterile distilled water, each vial contains 50 g of protein in 0.05 M Tris, 0.5 M NaCl at pH 8.0 with 1.25% trehalose.
