Experiment 1 quick description and guaiacol safety

observing the effects of different conditions such as pH, temperature concentration etc; on the rate of catabolic breakdown of peroxide by the enzyme peroxidase from turnips. RH2 + H2O2 -----------> R+ 2 H2O reducing agent = guaiacol

guaiacol Analysis: guaiacol is a colourless solution in it's normal state, but changes colour when it is oxidised. colour can measured using a spectrophotometer. Safety: eyes; guaiacol is an irritant and will cause redness and pain. [wear goggles] skin; causes irritation to skin. [ensure sleeves are completely down, wear gloves and wear suitable shoes] ingestion; Guaiacol is harmful if ingested. [do not drink, make sure to not touch your mouth after handling guaiacol and wash hands if necessary] inhalation; can be dangerous if inhaled. [presumably use a fume cupboard]

Much like guaiacol Hydrogen peroxide is an irritant, so protective eyegear, handgear and clothing must be warn, and care must be taken not to ingest or spill during handling.

Experiment 1 method (finding the enzyme baseline)

Equipment required: fill 3 test tubes with the reagents shown above and label them, these will be Pipettes (10ml & 5ml& stock reagents, place them in a test tube rack. Fill a small beaker with distilled 1ml ) water, and then label a fourth test tube reaction mix. Pipette tips (10ml & 5ml& 1ml ) label 3 test tubes 1 2 and 3 and and fill them with the reagents shown in the 7 test tubes table in the handout. 1 small beaker A test tube rack transfer some solution from tube 1 into a cuvette and zero the bottle of distilled water spectrophotometer. ( we use a reagent blank as, although the reagent may cuvettes look clear it may be slightly tinted and will therefore have a different absorption to clear water, i.e. it prevents zero error of the spectrophotometer. reagents required: mix tubes 2 and 3 to make the sample mixture and quickly add some to a clean H202 0.1% concentration (3mls) cuvette this will be t = 0 then record the absorption at 500 nm every 20 seconds until 2 minutes is reached. guaiacol (3mls) turnip extract (5mls) then plot a graph of absorption against time.
