Salmonella (except arizonae) XLD - pink colony with black centre. (typhi no black centre) DCA - colourless colony with black centre) Remember to plate the broth. API 20E Serology (cat 3)
Proteus Swarming colony API 20E spot indole (optional) Mirabilis - indole neg
Morganella, Providentia, Hafnia API 20E Hafnia can produce colourless colony on CTSMAC.
Serratia May produce red colony (marcescens) API 20E Lactose fermenter
API 20E
Fine GNR
Haemophilus (except H pittmaniae) Greyish flat colony - poor/no growth on blood agar, grows on chocolate agar X&V plate API NH Nitrocefin
X only - ducreyi V only - parainfluenzae, parahaemolyticus, paraaphrophilus (all para) X&V - influenzae, heamolyticus
Salmonella serology common orgs S typhi - O9, D, Vi S paratyphi A - O2 S paratyphi B - O4, b S paratyphi C - O6, O7, Vi
S enteritidis - O9, g S typhimurium - O4, i S montevideo - O6, O7, g S virchow - O6, O7 S dublin - O9, g
Do pan O and H, then test O4, O9 and g, i (as typhimurium and enteritidis are most common) if no result try others
Aeromonas Coliform, could be NLF or LF, could be haemolytic or nonhaemolytic API 20NE
Aggregatibacter actinomycetemcommitans GNCB, oxidase could be variable. Prolonged incubation needed. (Ask how long incubated for) pinpoint colony after 24hr. Firm adherent star shaped colony, non-motile, indole neg, urease neg Dont need X/V. Do not grow on Mac. Endocarditis.
Haemophilus (all except ducreyi) X&V API NH Nitrocefin Urease (Pittmaniae neg)
Capnocytophaga Fusiform shaped rod. Yellowish colony, spreading or gliding edge, human/animal bite. Aminoglycoside R
Campylobacter Microaerophilic, 42 deg C, seagull shaped bacteria in gram stain use carbol fuschine as counterstain/if strongly suspecting just put g. violet, wash, iodine, wash and look for the shape. Typical shape and oxidase + is enough
Gonococcus Presumptive diag = growth on selective media + appropriate colonial morphology + typical GN diplococci (long axis parallel) + Oxidase positive. Medicolegal - presumptive + 3 tests (biochemical, immunological and molecular - ref lab). High risk population (GUM) - Presumptive + 1 or 2 tests, Low risk population (GP) - presumptive + 2 tests,
