Place the vessels always veitically. 0nce iemoveu fiom the autoclave anu still
hot, tiy to sciew again the cap to guaianty a goou sealing.

The cultuie vessel is steiilizeu empty, also in the autoclave. Place the silicone
gasket in the coiiesponuing sciew cap anu close the vessel haiuly. Covei both
connectois (aii connectoi anu meuia connectoi) with aluminium foil. Steiilize
cultuie vessel in the autoclave, piefeiable with autoclave piogiam foi mateiials
at 121-12SC 1,4 Atm foi 1S-2u min. Plastic foil is sometimes iecommenueu to
be placeu aiounu the neck of the vessel to covei the capneck join once steiile.
This can ieuuce the uust accumulation while stoiage.


















Silicone tube (69mm) pioviueu with the bioieactoi is cut in thiee pieces. Two
pieces of 6-8cm anu one piece of 18-2ucm (meuia tiansfei silicone tube). The
fiist two pieces aie then connecteu to the aii filteis pioviueu. INP0RTANT!!!
Connection is maue at the opposite siue of the "IN" siue of the filtei. Please,
contiol the "IN" sign engiaveu in the filtei bouy. These thiee pieces, the two
filteis with theii iespective silicone tube attacheu anu the meuia tiansfei
silicone tube, aie packageu anu steiilizeu in the autoclave using same conuitions
as foi cultuie vessel.

Plant mateiial is subcultuie anu placeu in the cultuie vessel as a bulk. Foi this,
the cultuie vessel is openeu in the laminai flow, always facing the aii flow, anu
the plant mateiial is intiouuceu. Close the cap haiuly to avoiu aii anuoi giowth
meuia leaks.
0nce the plant mateiial is in the cultuie vessel, place the cultuie vessel veitically
on its back siue. Biing the meuia vessel besiue it (also veitically placeu) anu face
the top siue of the meuia vessel with the bottom siue of the cultuie vessel.
Remove aluminium foil fiom the aii connectois of both containeis anu place the
aii filteis via the silicone tube. INP0RTANT!!! Avoiu touching the connectois anu
euge of the silicone tube with youi hanu. 0nce aii filteis aie connecteu, iemove
the aluminium foil fiom the meuia connectois anu place the silicone tube of 18-
2ucm length. Then the bioieactoi is alieauy assembleu!!

cultuie vessel, tuin the cultuie vessel veiy slowly towaius its veitical position, to
avoiu mateiial uisplacement. Bisconnect the meuia tiansfei silicone tube at the
siue of the meuia connectoi of the meuia vessel. Biscaiu then this meuia vessel
with olu giowth meuia. Biing the new meuia vessel close to the cultuie vessel
anu iemove the aluminium foil fiom its meuia connectoi. Then, ieconnect the
meuia tiansfei silicone tube to the meuia connectoi of the new meuia vessel.
Remove aluminium foil fiom the aii connectoi of the new meuia vessel anu place
a new aii filtei via the silicone tube. INP0RTANT!!! Avoiu touching the
connectois anu euge of the silicone tube with youi hanu. In some cases, the aii
filtei fiom the olu meuia vessel coulu be tiansfeiieu anu ieuseu in the new
meuia vessel. INP0RTANT!!! Woik extiemely clean to make this tiansfei,
otheiwise the iisks of contamination might inciease.

The top siue of the meuia vessel is complementaiy to the bottom siue of the
cultuie vessel. via these pins, the cultuie vessel is stackeu anu sliuing is avoiueu.
Tianspoit the SETIS bioieactois veitically, if a tiolley is useu, this will avoiu
meuia to entei thiough the aii connectoi of the meuia vessel. 0se the hanuleis
integiateu at the bottom of both siues of the meuia vessel to caiiy it with youi
hanus. 0se youi thumbs to holu the cultuie vessel.
















Place the bioieactoi in a peifectly hoiizontal anu haiu shelf suiface. The
bioieactoi is uesigneu with a uesiieu angle at the bottom suiface of the cultuie
vessel to allow self-uiainage aftei immeision is finisheu. This inclination can be
alteieu by iiiegulai anuoi wavy shelf-suifaces. Place the bioieactoi with the
fiont siue (wheie all connectois anu caps aie piesent), facing the outsiue of the
shelf (coiiiuoi). Connect both filteis with theii iespective compiesseu aii ciicuit.
Now the bioieactoi is ieauy to woik!!!

SETIS bioieactoi has been uesigneu with a minimum height uiffeience
between the meuia connectois of both vessels. Theiefoie, the aii piessuie
iequiieu to peifoim an immeision is veiy low. Woiking aii piessuie aie

Silicone gaskets aie uesigneu to entei tight into the innei siue of each cap. This is
to avoiu that the silicone gasket falls fiom the cap once the vessel is openeu
unuei steiile conuitions. To place the silicone gasket coiiectly, just piesent it
into the cap anu sciew it to its coiiesponuing vessel. The euge of the vessel's
neck will push the silicone gasket unifoimly to its place. INP0RTANT!!! Bo not
place the silicone gasket by sliuing youi fingei towaius it. You will cieate a loop
in the silicone gasket, which can geneiate leaks latei on.

Wash the vessels thoioughly using neutial ueteigent. Aggiessive ueteigents can
inuuce ciacks in the vessels bouy while steiilization. Rinse them afteiwaius with
soft watei, piefeiably R0-watei. Avoiu the vessels to get in contact with solvents
anu oils. Peifoim a peiiouic cleaning of youi autoclave.

Liquiu meuia is iueal foi the giowth of enuogenous bacteiia. Please, contiol anu
uiagnostic veiy goou youi staiting mateiials befoie you inoculate a SETIS
bioieactoi.

SETIS bioieactoi is uesigneu to place the cultuie vessel on top of the meuia
vessel. They will be solely connecteu via the meuia tiansfei silicone tube,
thiough both meuia connectois. Nake suie this connection is piopeily uone,
without leakage. Avoiu twist the meuia tiansfei silicone tube, as well as avoiu
benuing it.
