Enzyme Lab Title: Enzyme-Controlled Reactions Problem/Purpose: Determine the effects of substrate concentration and pH on the initial rate

of an enzyme-catalyzed reaction. Hypothesis: If it affects the total amount of the data then the data want be right because you cant graph them or make charts.

Materials: 1. Computer 2. Website

Procedures: 1. pH level is adjusted by clicking the up or down arrows below the corresponding test tube. 2. To add substrate, click and drag the weighing paper to the appropriate test tube. 3.For all trails, assume the enzyme concentration in each test tube is constant (the same for each).

At test tube 1 and 0.5, pH 7 it was 72. And at test tube 2 and 1.0, pH 7 it was 145 and then for test tube 3 and 2.0, pH 7 it was 300. And then it changed to test tube 4 and 4.0, pH 7 it was 350 And then for test tube 5 and 8.0, pH 7 it was 350 again. Then for pH 3, 0.5 it was 19 then for 1.0 pH 3 it was 39. Then for 2.0 pH 3 it was 82 and then for 4.0 pH 3 it was 96. Last for 8.0 pH 3 it was 96. The last column which is pH 11 0.5 was 24 and then 1.0 for pH 11 it was 49 then for 2.0 pH 11 it was 103. Next for 4.0 pH 11 it was 121. Last it was 8.0 pH 11 it was 121 again.

In test tube 1 in the pH 3, 0.5g had 19 then in test tube 2 in the pH 5 column it was 39. Next in test tube 3 in the pH 7 it was 72. Then in test tube 4 pH 9 it was 45 and next test tube 5 pH 11 it was 24. In 2.0g test tube 1 pH 3 it was 82 and then in test tube 2 in the pH 5 column it was 168. Next in test tube 3 pH 7 it was 300. And then in test tube 4 pH 9 it was 189, after that it was test tube 5 pH 11 it was 103. Then in 8.0 test tube 1 pH 3 it was 96, than in test tube 2 pH 5 it was 198 and next after that in test tube 3 pH 7 it was 350. After that in test tube 4 pH 9 it was 223. Last in test tube 5 pH 11 it was 121.

Conclusion: As you can see the graph werent that similar but there was some number that was on graph 2 that was on graph 1 and the other way around. I felt that this lab was kinda of hard but it was fun doing the lab on the computer. What we had to do in this lab was determine the effects of substrate concentration and pH on the initial rate of an enzyme-catalyzed reaction.
