Aflatoxin M1

Aflatoxin is a toxic and carcinogenic substance produced by certain strains of the


moulds Aspergillus flavus and A. parasiticus. When ruminants consume grain containing
aflatoxin, they create a metabolite which is found in their milk, aflatoxin M1.
Ingesting excessive amounts of the toxin can cause chronic health and performance
problems or death in animals. Aflatoxin has been shown to cause liver damage or
cancer, decreased milk and egg production, immune system suppression and
interference with reproductive efficiency.

Legislation
The EU has set a maximum residue limit for aflatoxin M1 of 50 parts per
trillion (ppt) in raw milk, heat-treated milk and milk for the manufacture of
milk based products. There is also an EU limit of 25 ppt for Infant formula
milk and follow on formula milk as well as dietary foods for special medical
purposes intended specifically for infants.

Procedure

Note: Please read kit instructions completely before performing test.

1. Add 250 L of controls and samples to their respective red-marked


mixing wells.
2. Transfer 100 L from the mixing wells to the antibody coated wells.
3. Place microwell holder on an automatic plate shaker and allow
continuous shaking for 20 minutes at room temperature.
4. Dump liquid from antibody wells. Wash wells thoroughly with diluted wash
buffer, repeat wash step 5 times. Tap out liquid on absorbent paper.
5. Transfer 100 L of conjugate from the reagent boat to the wells.
6. Place microwell holder on an automatic plate shaker and allow
continuous shaking for 10 minutes at room temperature.
7. Dump liquid from antibody wells. Wash wells thoroughly with diluted wash
buffer, repeat wash step 5 times. Tap out liquid on absorbent paper.
8. Transfer 100 L of substrate from the reagent boat to the antibody wells.
9. Place microwell holder on an automatic plate shaker and allow
continuous shaking for 15 minutes at room temperature.
10. Transfer 100 L of red stop solution from the reagent boat to the wells.
Mix by sliding microwell holder back and forth on a flat surface.
11. Read results using a microwell reader with a 650 nm filter.
