Hello saimeghana,
Regarding you first query.."main difference between main work center and work center plant in order."
Main work center is the responsible work center for the order. This is the work center responsible for the technical object on which order has been created.
Work centers assigned in the operation are the work centers responsible for that particular operation. It may be same as main work center or you can assign
operations to other work centers also.
Similarly responsible cost center is responsible for execution of the order. This the the cost center assigned to main work center.Order never gets settled on this
cost center.
Another cost center in the location tab is the cost center (You can call it requesting cost center) assigned to the technical object on which the order has been
created.
And normally orders get settled on this cost center(which is assigned to the technical object).

From SAP Help :
The cost center "printing works" requires a new machine for folding newspapers. The cost center "site office" is responsible for implementing the construction.
The cost center "site office" is the responsible cost center.
The cost center "printing works" is the requesting cost center.

deepak

Main work center => Responsible for all the tasks in the order
Work Center/Plant => Executing work center for each operation
Responsible Cost center => Labor cost center (for which activity pricing will be done with reference to activity types)
Cost Center in Location tab => Department / Cost object which uses this equipment (Settlement cost receiver object)
