CULTURE Inoculate stool sample on Selenite F broth and Alkaline Peptone water then incubate it at 37 C for 10-12 hours
Subculture from Selenite F to XLD agar
Subculture from Alkaline Peptone water to TCBS Incubate agar plate at 37 C for 24 hours
For Salmonella and Shigella- pick one colony of each
morphologic type of red to red orange colony and any colony with black centre from XLD . Subculture each colony picked to TSI and Urea agar. If urease test positive (pink) discard the sample, if negative proceed to TSI agar-if TSI agar shows acid butt(yellow) and alkaline slant (red) proceed to next step otherwise discard the sample Confirm Salmonella and Shigella organism by API 20E Test for agglutination for salmonella and shigella (Serology) For Cholera- pick Yellow colored colonies from TCBS are taken and confirmed by API 20E Oxidase and Cholera red reaction is also done to identify Vibrio
