YEPD AGAR (7294)

Intended Use
YEPD Agar is used for the maintenance and propagation of yeasts.
Product Summary and Explanation
YEPD Agar, Yeast Extract Peptone Dextrose Agar, is used often in yeast genetics studies for cultivating
Saccharomyces cerevisiae and other yeasts.1 Yeasts grow well on a minimal medium containing only
dextrose and salts. The additional source of nitrogen and vitamins permits faster growth of yeasts.
YEPD Agar is also referred to as YPD Agar, and is used extensively in molecular microbiology procedures.2,3
Principles of the Procedure
Peptone provides the nitrogen and amino acids in YEPD Agar. Yeast Extract supplies vitamins and minerals
to stimulate growth. Dextrose is the carbon energy source. Agar is the solidifying agent.

Precaution
1. For Laboratory Use.
Directions
1. Dissolve 65 g of the medium in one liter of purified water.
2. Heat with frequent agitation and boil for one minute to completely dissolve the medium.
3. Autoclave at 121C for 15 minutes.
Quality Control Specifications
Dehydrated Appearance: Powder is homogeneous, free flowing, and beige to yellow-beige.
Prepared Appearance: Prepared medium is trace to slightly hazy, and light to yellow beige.
Expected Cultural Response: Cultural response on YEPD Agar at 25 - 30C and examined for growth at
18 72 hours of incubation.

Results
After sufficient incubation, the medium should show growth as evidenced by formation of isolated colonies
and/or a confluent lawn of growth on the surface of the agar.
Storage
Store sealed bottle containing the dehydrated medium at 2 - 30C. Once opened and recapped, place
container in a low humidity environment at the same storage temperature. Protect from moisture and light by
keeping container tightly closed.
Expiration
Refer to expiration date stamped on the container. The dehydrated medium should be discarded if not free
flowing, or if the appearance has changed from the original color. Expiry applies to medium in its intact
container when stored as directed.
Limitation of the Procedure
1. Due to nutritional variation, some strains may be encountered that grow poorly or fail to grow on this
medium.
2. If adding supplements such as amino acid mixtures, care should be taken to avoid agar hydrolysis. Add
the supplements after autoclaving and do not re-melt.
