PRINCIPLE

The DENSIMAT is a densitometer used to measure the density of a bacterial


suspension.
An incident beam of light is passed through the ampoule and two
subsequent measurements are obtained:
- Scattered light S (two photosensors)
- Transmitted light T (one photosensor)

The ratio S/T is directly proportional to the density of the bacterial


suspension.
The results are displayed on a mobile graduated scale on an LCD (Liquid
Crystal Display). The number of squares displayed is proportional to the
McFarland value.

Before Opening the Boxes:

l Make sure that no damage has been caused during transportation.


If damage has occurred, make a claim to the transport company and notify
Customer Service at bioMrieux SA.

When Opening the Boxes:

l Make sure that there a11 the items listed on the packing list are included in
the boxes. If possible keep packaging materials in case the system has to
be moved in the future.

GENERAL DESCRIPTION

The real value of the bacterial density cornes from the calculation of the
ratio between the scattered light and transmitted light of the suspension.
The light source is an infrared source contained in the instrument.
The electronics of the DENSIMAT module are made up of two electronic
cards. These allow the space in the instrument to be optimized and possible
hanger-on with the signal to be decreased.

SENSOR BOARD
DESCRIPTION
The sensor board allows conversion of the light received on the
photosensors in proportional analogie signals.

- Preamplifier of Transmitted Light


The preamplifier circuit converts the current from the photosensor to a
voltage proportional to the ratio below.
Ratio = 100 mV / pA

- Preamplifier of Scattered Light


The preamplifier circuit converts the current from the photosensors to a
voltage proportional to the ratio below.
Ratio = 270 mv / pA

- Light Source Power Supply


The LED used as a power source should be power supplied with a constant
current.
For the components with U2, the constant current should be 76 mA
The switching ON/OFF of the LED is controlled by the logical level on the
3/U2 pin. The LED is ON when the logical level is 0.
The C7 capacitor decreases the stabilization time of the current in the
circuit.

- Power Supply Micro Switch


The SWl micro switch is an interrupt with contact no gift.
The plug for the external power supply is a coaxial plug with a BREAKBEFORE-MAKE switch to avoid loading the battery with current.
On the battery line the Dl Schottky diode limits the voltage on this line.
The Fl fuse (750mA 125V) protects the battery set.
The C6 capacitor is used as a regulator when the LED is switched ON.

- Optical Interrupt
This optical interrupt is used to identify the typeof ampoule when it is put
into the optical block.
This function is done with the OP1 optical interrupt.
TO reduce consumption, the optical interrupt uses an analogie OUTPUT.

- Processing of Transmitted Light
The processing of the signal cornes directly from the SENSOR board.
The processing is done through the U3 circuit section C. The amplifier
circuit is a noninverting operational amplifier.
The bandwidth is limited with a continuous theorical gain equal to 23.
The R23 resistor limits the current through the INPUT of the A/D
converter included in the microcontroller.
The Dl Schottky diode limits the voltage through the INPUT of the A/D
converter to VCC + 0.5V. It is necessary to avoid wrong converting of the
channel next to the converter, to make sure the voltage through the
INPUT does not exceed the reference voltage by more than one volt.
The signal of transmitted light goes directly to an INPUT of the A/D
converter through the R24 resistor, which limits the input current.
In this case, the Schottky diode is not required to protect the INPUT,
because the gain is 1 and it cannot exceed the reference voltage.

- Processing of Scattered Light


Processing of the signal cornes directly from the SENSOR board.

The processing is done through the U3 circuit section B. The amplifier


circuit is a noninverting operational amplifier .

The R28 resistor limits the current through the INPUT of the A/D
converter included in the microcontroller.
The D2 Schottky diode limits the voltage through the INPUT of the A/D
converter to VCC + 0.5V. It is necessary to avoid wrong converting of the
channel next to the converter, to make sure the voltage through the
INPUT does not exceed the reference voltage by more than one volt.
Processing is done through the U3 circuit section A. The amplifier circuit

- Processing of the Battery Source


This function allows you to check the battery voltage and display the level
of the battery.
Processing is done through the U3 circuit section D. The voltage of the
battery is divided between the two resistors R6 and R7 (7.2V nominal) to
adapt it to the characteristics of the A/D converter. The OUTPUT of the
amplifier serves as a buffer and the signal goes directly to an INPUT of the
A/D converter through the R29 resistor, which limits the input current.
If the battery voltage goes below 5.76 V the battery change indicator light
cornes on on the display.

-Management of the DENSIMAT


Management of the instrument is controlled by a single chip Ul
microcontroller. This circuit allows the following functions:
- Hardware control.
-Conversion of the analog signal from sensors into a numerical
signal.
- Reading management.
- Calculation and displaying of the result.
The microcontroller used is the single chip MC68HC711E9 from
MOTOROLA. Its characteristics are:

Values of calibration

With this microcontroller, the U2 circuit and the Cl4 Power-on Reset
capacitor allow the power supply level to be monitored. When the voltage
goes below 4.3V, the system generates a general reset.
Connexion with the LCD circuit is done through port B.
The high level of the PB3 line allows you to switch ON the light source
(LED).
The high level of the PB4 line allows you to manage the optical interrupt of
the type of ampoule.
The PAO line is used to read and manage information from the sensors.

- Management with an External Computer


Management with an external computer is done through the 52 connecter
Lines PB5 and PB6 are used during the processing of the adjustment.

- Liquid Cristal Display


The standard LCD component uses 16 ASCII characters per line on two
lines.
The LCD brightness is fixed by resistors R20 and R31.
The interface with the microcontroller is done to 8-bit MPU through PIA
(Port C) Signals PBO (Enable), PBl (Selects Read Write) and PB2 (Selects
Register) are used to control the OUTPUT.

- Power Supply of the Numerical Part


The power supply voltage VCC is produced using the U7 circuit and the
supply voltage obtained (5 VCC) is produced by the battery voltage.

- Power Supply of the Analog Part


The A/D converter circuit requires a voltage of 5000 mV.
This supply voltage is obtained from the U6 circuit.
The operational amplifiers require voltages at + 8 V ( +V) and -8 V C-V).
The supply voltage +V is obtained from the U4 circuit through the U6
circuit.
The supply voltage -V is obtained from the U5 circuit through the U4
circuit.

DESCRIPTION
The optical block is a mechanical block, where the LEDs and photosensors
are positioned to make nephelometric readings of an ampoule.
The LED is SFH400 III.

Preliminary Procedures
1) Make sure that the optical block is clean.
2) Check the external appearance of the DENSIMAT to detect any
damage.

Scheduled Procedures

Materials used:

Clean the photosensors.


Using the cleaning tool and a sterile swab dampened with
alcohol, carefully clean the photosensors.

Check the calibration.


Using ampoules from the kit, check the values obtained with
the DENSIMAT for each type of ampoule (large and small) and
compare them with values written on the ampoules.

CAUTION!

Depending on the results you should perform the


DENSIMAT calibration procedure.

Using the cleaning tool and a sterile swab dampened with alcohol,
carefully clean the photosensors located in the optical block.

Additional Cleaning Procedure


The following cleaning procedure is not included in the monthly cleaning
procedure. It is the operators responsability to decide how often it should
be performed.
External Surfaces
The external surfaces of the DENSIMAT should be cleaned with warm soapy
water to prevent the build-up of dust.

Glossary

LED 950 nm

Light Emitting Diode 950 nm.


Light source emitting on the 950 nm wavelength.

Nephelometry

Measurement of the intensity of light scattered (S) at 30,


directly proportional to the bacterial growth.

McFarland

Standardization unit for measurement of bacterial


density.

Optical density

The result of the measurement of a medium through an


incident beam of light.

Photosensor

Sensor enabling a light source received to be transformed


into a source of electrical current.

Scattered Light

Light which is spread by the bacteria. In this case, the


light cornes out of the ampoule in a direction which is 30
from the direction of the emitted light.

Transmitted Light

Light which passes through the ampoule in the same


direction as emitted light.
