PROCEDURE

1. The wastewater sample is collected.

2. For influent sample, a volume of sample is pipetted (20mL) and poured into each
6 BOD bottle
3. Nitrification inhibitor is poured into 3 BOD bottle.
4. Aerated distilled water is poured inside the 6 BOD bottle to top it up until the BOD
bottle neck.
5. After that, the dissolved oxygen (DO) of all the samples is measured by using the
DO meter. And the reading is recorded as the initial DO of the samples.
6. All the samples is closed with the BOD bottles cap and wrapped up with the
aluminium foil to ensure no oxygen can penetrate into the bottles and then, all
the bottles is placed inside an incubator to incubate it at the temperature of 20 oC
for 5 days.
7. On the 5th day of the incubation, all the samples is measured again by using the
DO meter to get the final DO and measure the BOD5 of the sample.
8. Step 1-7 is the same for effluent sample but with different volume pipetted
(150mL).
BOD test for a blank sample. All of the procedure is all the same as the above sample
except this time; the sample is the aerated distilled water itself which is to be filled
fully into the BOD bottle (300mL) and the student will come on 5 th day of the
incubation to get the final DO and measure the BOD5 of the sample.
BOD test for coke and milk sample. The procedure is also the same with the previous
BOD test except, the sample is diluted first with the ratio of 1:1000 for both coke and
milk. Then 20mL of each of the sample are taken and poured into three BOD bottles
for coke and another three BOD bottles for milk together with another 2mL of seed
(influent wastewater) for each of them and the student will come on 5 th day of the
incubation to get the final DO and measure the BOD5 of the sample.
BOD test for the standard and for the seed. For the standard, the procedure is the
same as previous, except, both 150mL of glutamic acid and 150mL of glucose is
mixed first inside a volumetric flask and 2mL of the mixture is the pipetted and
poured into each of three BOD bottles together with 2mL of seed. This student also
does the BOD test for the seed. The procedure is the same except, the sample; 2mL
of the seed (influent wastewater) is pipetted and poured into each of the BOD bottles
and the student will come on 5 th day of the incubation to get the final DO and
measure the BOD5 of the sample.
