centuriesintheHinduKushmountainrangesofpresentdayPakistan.PreviousYchromosomeand
mitochondrialDNAmarkersprovidednosupportfortheirclaimedGreekdescentfollowingAlexanderIIIof
Macedonsinvasionofthisregion,andanalysisofautosomallociprovidedevidenceofastronggenetic
bottleneck.Tounderstandtheiroriginsanddemographyfurther,wegenotyped23unrelatedKalashsamples
ontheIlluminaHumanOmni2.5M8BeadChipandsequencedonemaleindividualathighcoverageonan
IlluminaHiSeq2000.ComparisonwithpublisheddatafromancienthuntergatherersandEuropeanfarmers
showedthattheKalashsharegeneticdriftwiththePaleolithicSiberianhuntergatherersandmightrepresent
anextremelydriftedancientnorthernEurasianpopulationthatalsocontributedtoEuropeanandNearEastern
ancestry.SincethesplitfromotherSouthAsianpopulations,theKalashhavemaintainedalowlongterm
effectivepopulationsize(2,3192,603)andexperiencednodetectablegeneflowfromtheirgeographic
neighborsinPakistanorfromotherextantEurasianpopulations.Themeantimeofdivergencebetweenthe
Kalashandotherpopulationscurrentlyresidinginthisregionwasestimatedtobe11,800(95%confidence
interval=10,60012,600)yearsago,andthustheyrepresentpresentdaydescendantsofsomeofthe
earliestmigrantsintotheIndiansubcontinentfromWestAsia.

TheexaminationofDNAshowsthattheKalashcommunityhasmoreincommonwithSiberianhuntergatherersthan
AlexandertheGreat.
