C. WORK PROCEDURES
1. Mucin Test
Millon Benedict Mollish
Reagent Reagent Reagent

5 mL saliva 2 drops of Using water as a control by

adding 2 drops of 0.1M acetic Filter the precipitate
acetic acid
formed
0.1M acid 2 drops
Testing

2. Thiocyanate test
If red still means there thiocyanate, if
the red color is missing then no
thiocyanate

5 mL saliva 5 drops of FeCl 1 drop of Using water as a control by 5 drops of HgCl

0,1M concentrated adding 5 drops of FeCl 0.1M and 1%.
HCl 1 drop of concentrated HCl

3. Saliva Test Composer Inorganic Compounds

1 2 3 4

15 mL saliva
Drops with 2 N acetic acid,
forming a precipitate Enter into 4 different reaction
Heated to boiling filtered The filtrate tubes
was inserted
3 mL

Checking ion Cl

1 1

Acidified with Added a few

dilute HNO3 drops of 0.5 M
AgNO3

Checking PO43-
2 2

Acidified with Added 1 mole of reagent

dilute HNO3 ammonium molybdate be heated

Checking ion SO42-

3 3

Acidified with HNO3 (e) Added 1 mL CaCl2 2%

Checking ion Ca2+

Added 1 mL of NH4 - Oxalate 4%

In each experiment carried out observations of the results are instantaneous, and
also after one hour and then take a good look possibility precipitate formed little
that it is difficult to observe.

4. Effect of Temperature Tests Against Activities Ptiatin

2 1 2 3

Each tube was added Tube 1 be

cooled tube 2 at room
temperature To the tube was added
5 mL of 1% starch. saliva 1: 9
tube 3 heated to a
temperature of 38oC

Every 5 minutes to take samples and checked tube 4 added salivary that has
in with 2 drops of 0.01 M I2, and record the been heated for 5 minutes
speed of the starch solution in each tube

5. Test Estimation Ptialin

2 mL NaCl 0,1M

10 mL starch 1% Into a test tube inserted 8 3 moles of water and 3 drops of a

solution of 0.01 M I2 1 mL of saliva 1: 9 into a
tube containing starch
Heated at 38oC

Record the time at the time of adding any starch

mixture of saliva in a solution of I2
indeterminate cause the color
If the time it takes less than four minutes to do
the experiment under the same way but saliva
should be diluted 2 times

Intervals of 30 seconds with a pipette take the starch mixture

and insert 2 drops into the tube 8

6. Determination of pH tests from which to work saliva

Heated at 38oC.

Adding I2 and specify in which a tube is missing The addition of acetic acid
the color point is reached so that the mixture becomes
acidic atmosphere
