Background and objective:

Forensic science plays a vital role in solving criminal cases through


applications of different sciences. Among all of these,DNA profiling is most
promising and authenticated procedure,which basically utilize a specific part of
DNA called DNA markers. There are different DNA markers routinely used now
i.e STRs ,SNPs Amelognin,SRY etc from both autosomal and haploid cells. Mt
DNA has advantage on nuclear DNA. mt DNA is a maternally inherited
polymorphic and resistant to cellular degradation.it is important when there is very
low or little sample.mtDNA has number of advantages which make it valuable
marker for forensic DNA typing and population studies.Being in large number per
cell,maternal inheritance,resistant to

Sudozai tribe settled in different areas of Pakistan and especially in AJK, is traced
back to compaginin of prophet Mohammad (s.a.w) hazrat Qaees abdul
rasheed.Hazrat Qaees abdul rasheed himself belong to tribe of bani Israel. 9 th
descendant of Hazrat Qaees abdul rasheed was sudo bin atman was inhabitant of
Afghanistan and 17th descendant was jasi khan moved from afghanistan to
Pakistan(charsada) in 1284 A.D. In this study we aim to characterize the mtDNA
control region of sudozai population and to see the association with other Pakistani
population on genetic level.

New knowledge to be Gained:


1-Characterization of mt DNA of Sudozai population.

2.Genetic Structure Prediction of this population

3.Exploration the route and origin traced back (if any ) to bani-israel

4.Addition to mt DNA database of Pakistan.

Material and Methods:

(i) Subjects and Models:


Our study subjects will be unrelated males members of sudozai
population from different areas of Pakistan mainly from AJK.
(ii) Study Design:
Sampling will be done by extracted 3-5 ml of venous blood from 100
unrelated healthy males of sudozai across Pakistan.sampling will be
performed according to ethical disciquittes of voluntaries.
DNA EXTRACTION of all samples will be done by using phenol
chloroform method.
QUANTIFICATION of mtDNA will be performed by using NanoDrop
1000 spectrophotometer.
PCR Amplification of control region (16024-576) will be done by
designing primers from http://forensic.... Condition will be set by
troubleshooting.Then PCR products shall be sedquenced commercially
and ensure the DATA quality bidirectional strategy will be performed.
DATA ANYLISIS will be done using chromas lite version and sequence
scanner software2. Comparison of sequences will done with rCRS
(revised Cambridge Reference sequence)(S.Anderson et al 1981).mtDNA
profiler and CMMA will be used for haplogroup allocation.
Statistical Analysis. Simple calculation of counting haplogroups.Genetic
diversity, random match probability and power of discrimination.By
using arlequnin software (version 3.5) analysis of molecular variance
will done. DATA will also be compared with other population of Pakistan
and with jews population of world.
