These methods are based on the measurement of absorption, emission, or fluorescence originated
from the free, unionized atoms or atomic ions in gas phase.
In atomic absorption and fluorescence spectrometry the
atomizer should produce a large population of sample atoms that are in
the ground state. The sample matrix should not affect the degree of
atomisation, or otherwise chemical interferences are encountered.
Different combustion flames are most commonly used as atomization
devices. In addition, graphite furnaces and flame or electrically heated
quartz tubes are commonly used to enhance the determination of low
analyte levels by AAS. In AES the atomizer/excitation source must be
very efficient or otherwise only the elements having low lying excited
states can be measured with reasonable sensitivity (Figure 2). Various
plasma sources, especially ICPs, fulfil this well, especially when liquid
samples are used. The atomizatiodexcitation devices used in AAS, AES
and AFS techniques can be at the atomization temperature continuously
(flame, plasma) or transiently (graphite furnace)
RAMAN
Los espectros Raman se obtienen al irradiar una
muestra con una fuente potente de rayos laser de radiacion
monocromatica visible o infrarroja. Durante el
proceso se registra el espectro de la radiacion dispersada
a un cierto angulo, casi siempre 90°, con ayuda de
un espectrometro apropiado. Para evitar la fluorescencia,
las longitudes de onda de la excitacion se eliminan
de una banda de absorcion del analito. El experimento
Raman se ilustro ya en la figura 6.18. Las intensidades
de las lineas Raman son, cuando mucho, un 0.001% de
la intensidad de la fuente. Debido a esto podria parecer
mas dificil detectar y medir las bandas Raman que
las bandas vibracionales en el infrarrojo.
Sin embargo,
la radiacion Raman difundida esta en las regiones visible y del infrarrojo cercano, para las cuales ya
hay
detectores muy sensibles. En la actualidad, las mediciones
de los espectros Raman son casi tan faciles de
hacer como las de los espectros en el infrarrojo.
