Use of the Microscope

It is best to consult the manufacture’s manual before using the microscope because
it is the most sophisticated equipment of this type. Although it is essentially alike with
flourescence microscope, they may different in some points of operation. Some important
precautions that should be known, such as:

 Never inspect the mercury arc lamp while it is hot. When the lamp is hot, the
lamp’s pressure is increased, making the pressurized arc lamp become
potential bomb.
 Never expose your eyes to the direct rays of mercury arc lamp. Unfiltered
light from the lamp is rich in both ultraviolet and infrared that can damage
the eyes.
 Be sure that the barrier filter always in place when looking down through the
microscope. If the barrier filter is removed, your eyes can be damaged by
ultraviolet rays.

Question:

1. Why must you consult the manufacturer’s manual before using the microscope?
A : Because the microscope is the one of the most sophisticated equipment of
this type, i must consult the manual to understand the procedures and
precautions of the microscope.
2. What is the cause of microscope hazard?
A : Inspecting the arc lamp while it is hot, exposing eyes to the direct rays of the
arc lamp, and looking down to the microscope when the barrier filter is removed.
3. Why must you do not expose your eyes to the direct rays of mercury lamp?
A : Because if i exposing my eyes to the direct rays of mercury lamp, the light is
rich in both ultraviolet and infrared, which can damage my eyes.
4. What must you do before inspecting the mercury arc lamp?
A : I must let the lamp cool completely before inspecting it.

B. Putting a microscope away

When taking the microscope out from cabinet, you expecting it to be clean and in
proper condition. The next person after you have used the microscope will expecting the
same condition. Some of the procedures can ensure this conditions, such as:
 Remove the slide from the microscope stage,
 Wipe off the immersion oil from the lens and stage with lens tissue,
 Wrap the electrical cord around the base if the microscope has a long
attached cord,
 Wrap the dustcover.
Question :

1. Why should you keep the microscope clean and in proper condition?
A : Because if i keep the microscope clean and in proper condition, next person
to use the instrument do not have to clean it first.
2. Why should you wipe off the immersion oil from the lens and stage with lens
tissue?
A : Because the immersion oil could attract dust to the lens, which can distort the
objective lens.
3. Why should you wrap the microscope with dustcover?
A : Because the dustcover can protect the microscope from dust and keep the
microscope clean.

C. The Liquid Filled Thermometer

The type of thermometer depends of a liquid associated with an increase in

temperature. The most common type is mercury-in-glass thermometer. On heating, the
mercury expands relative to the glass container and column is pushed along the bore of
the tube. The scale along the tube are calibrated in units of temperature. The scale gives
direct reading of temperature. Although it is accurate, the mercury-in-glass
thermometer is very fragile. This type of thermometer should not be used in food
industry because mercury poisoning could occur if it is broke.
The mercury fluid may be replaced by other fluids according to the application.
Mercury-in-glass thermometer is stable and has accurate performance, that is why it is
supplied with Temperature Measurement Bench.

Question :
1. What is the function of thermometer?
A : The function of thermometer is used to measure temperature.
2. How does the thermometer works?
A : When the thermometer’s sensor tip is close to the object, the liquid inside
thermometer will expanding until it is stopped at a specific scale of temperature
unit.
3. What is the most common type of thermometer?
A : The most common type of thermometer is mercury-in-glass thermometer.
4. Why mercury-in-glass is the most common type of thermometer?
A : Because the mercury liquid inside the thermometer has stable and accurate
performance.
