MID TERM SEMESTER EXAMINATION-2016

M. Sc. II Semester
Botany and Zoology
Paper-Advanced Molecular Biology
Time: 1 hour Max marks: 20

Note: The question paper is divided into two sections. Section A has short answer type
questions (not less than 125 words) while section B has long answer type questions. Choose
any four questions from section A and one question from section B.

Section A (2x4=8)
1. Identify the following structures.

(A) (B) (C)

2. What is Cot curve and its significance?

3. In the diagram below the light lines indicate the template strands and the dark lines
indicate new complementary strands.
A. Indicate where the replication origin was located with an ‘X’.
B. Label the 5´ and 3´ends of the new strands.
C. Label the leading-strand and lagging-strands.

D. Add arrowheads to the dark lines to show the direction in which the newly made
DNA strands were synthesized.

E. Number the Okazaki fragments on each strand 1, 2, and 3 in the order in which they
were synthesized (1 first, 3 most recently).

F. Indicate the direction of movement of the replication forks with arrows.

4. Give the name for A,B,C,D,E,F,G,H,I in the following picture?

5. How can E. coli divide faster that the time it takes to replicate it's chromosome?

Section B

Unit I

6. Write the name of different components of nucleic acid. Describe structure of a DNA
molecule.

7. What is Cot curve? How is it interpreted?

Unit II (1x12=12)

8. How will you justify that DNA follow semi conservative mode of replication? How a
bacterial DNA start to replicate?
9. What are the main DNA damaging agents? Describe base excision and nucleotide excision
repair method.
