This
protocol
contains
recommended
methods
for
evaluating
biomethane
production,
but
McGill
University,
Anaergia
and
UNU-‐
INWEH
bear
no
responsibility
for
the
health
and
safety
of
individuals
using
these
methods
in
a
laboratory
setting
outside
our
institutions.
It
is
recommended
to
wear
the
appropriate
personal
safety
equipment,
to
work
in
a
chemical
fume
hood
whenever
possible
and
to
autoclave
biological
waste
prior
to
disposal.

Weigh
out
the
equivalent
of
20
g
VS-‐inoculum
(to
obtain
80
g
VS-‐inoculum/L
of
working
digester
volume)
and
add
to
digester
(500
mL
Wheaton
bottles).
If
needed,
use
a
small
amount
of
distilled
water
to
transfer
sample
into
digester.
Store
in
fridge
until
ready
to
use.
2. Weigh
out
4
g
VS-‐waste
(to
obtain
0.2
g
VS-‐waste/g
VS-‐inoculum)
and
homogenize
with
100
mL
water
in
blender.
3. Add
homogenized
waste
to
digester
and
raise
overall
volume
to
the
desired
working
volume
(250
mL)
with
distilled
water.
4. Purge
digester
by
bubbling
CO2
or
a
mixture
of
N2/CO2
(80/20,
v:v)
in
the
liquid
mixture
for
5-‐10
minutes.
Optional
step:
Insert
two
needles
into
the
butyl
rubber
stoppers
of
the
cap,
one
feeding
CO2,
the
other
allowing
the
gas
to
escape
the
bottle).
Slowly
place
the
cap
on
the
bottle
while
removing
the
bubbling
purge
from
the
bottle
and
quickly
screw
the
cap.
Continue
purging
through
the
needle
for
5
minutes
and
then
remove
both
needles
from
the
rubber
cap.
Ensure
this
method
did
not
create
a
pressure
build
up
in
the
bottle
(FIGURE
4).
5. Incubate
in
water
bath
at
35°C
with
the
water
level
to
the
bottle
neck.
6. After
a
30-‐minute
equilibration
period,
using
the
syringe,
purge
digesters
of
any
pressure
build
up.
7. Shake
each
bottle
for
1
minute
every
day
(keeping
the
bottle
in
the
water).
