Procedures:

1. Label four boiling tubes as A, B, C and D. [ ]

2. Record the time taken for the lime water to turn cloudy in a table. [ ]

3. Place the boiling tube in a water bath of temperature 35oC. [ ]

4. Filled each boiling tubes as given below. [ ]

Boiling Tube Contents

A 10ml of yeast suspension + 10ml distilled water + paraffin oil
B 10ml of yeast suspension + 10ml of 5% boiled glucose solution + paraffin oil
C 10ml of yeast suspension + 10ml 10% boiled glucose solution + paraffin oil
D 10ml of yeast suspension + 10ml 15% boiled glucose solution + paraffin oil
4. Connect the boling tube to a test tube containing 2ml of lime water via a delivery tube.[ ]

5. Repeat the experiment for three times to get average result. [ ]

Tabulations of data:

Boiling tube Concentration of glucose/% Time taken for the lime water to turn cloudy/minutes
A 0
B 5
C 10
D 15
Discussion:

Based on the result of the experiment, the time taken for the lime water to turn cloudy is the shortest among the other
concentration is _____________________________________________which is only _________ minutes. Then is
followed by __________________________________ and ___________________________________. This is because
when the concentration of nutrients increased, the activity of yeast also increased. However, the lime water of boiling
tube A remain clear because there is no activity of yeast when there is no nutrients.

In this experiment, the anaerobic respiration condition is maintained by using boiled water to prepare the glucose
solution and yeast suspension. A layer of paraffin oil is added on the top of the reacting mixture to prevent atmospheric
oxygen from entering the mixture. The lime water turns milky because carbon dioxide gas is produced during anaerobic
respiration of yeast.
