GENERAL INSTRUCTIONS TO STUDENTS:

1. Students are reminded that practical work is the foundation on which the study
of sciences stands and every student is required to give the most earnest and
careful attention to it.
2. The laboratory work is arranged to accompany as closely as possible the
principles of theory discussed in the lecture classes.
3. Students must observe strict silence in the laboratory.
4. Eating, chewing, drinking, smoking or application of cosmetics is expressly
forbidden in laboratory areas. Storage of food items and outdoor clothing is also
not permitted.
5. No outdoor clothes are to be brought into laboratory areas.
6. Laboratory coats (properly fastened) must be worn at all times in all laboratory
areas including instrument rooms.
7. Place all extra clothing, unnecessary books, backpacks and purses in an
appropriate place. The laboratory area must be kept free of articles not in use.
8. Always keep the containers of alcohol away from open flames.
9. All cuts and abrasions should be covered with a waterproof dressing before
beginning any laboratory work.
10. Disposable protective gloves should generally be worn as necessary (and
changed regularly!) When working at the bench- particularly when handling
chemicals, or any biological material likely to contain micro-organisms.
11. No one is admitted to the lab room 10 minutes after the lab begins.
12. Each student must have his/her own lab record book/Journal.
13. The pages of the notebook/Journal must be numbered in pen all the way to the
end. This is to ensure that none are removed during the year.
14. Lab reports in the notebook are due at the end of the lab period.
15. Anyone caught plagiarizing work in the laboratory notebook, from a current or
past student's notebook, will be liable to punishment.

16. Students are responsible for the apparatus which are placed for their use on the
work bench.
17. If any article is stolen by the student, he/she will be suspended from the
laboratory.
18. Students breaking articles or apparatus will be required to replace it.
If the articles are broken on account of carelessness, they will have
to pay a fine in addition.
19. Cost of breakages should be paid within seven days.
20. When they finish their work they must leave the apparatus in the same position
in which they were placed originally.
21. On completion of experimental work, the workstation and all equipment must
be thoroughly cleaned.
22. All laboratory personnel should wash their hands at regular intervals and always
immediately when contamination is suspected. Hands should always be
thoroughly washed and rinsed on completion of all experimental work and
always before leaving the laboratory.
23. All solid and filter paper waste should be in the dustbin.
24. Lone working or carrying out experimental work in departmental research
laboratories outside normal working hours is not permitted other than by prior
arrangement.
25. Before leaving the lab be sure all electrical devices are unplugged from the wall.
26. Any faults to items of laboratory equipment should immediately be reported to
On Duty staff so that appropriate action can be taken. Prompt notification can
then be given to other potential users and if necessary the equipment in question
can be removed from service until repairs are carried out.
27. Report all accidents and injuries immediately to the instructor.
28. The certificate for the practical course required by the University regulations
will not be granted unless the student's progress has been satisfactory. This will
be judged not only by the practical work done in the laboratory, but also by the
neat, systematic, regular writing of the accounts in the home record book.

INSTRUCTIONS TO DEPARTMENTAL LABORATORY PERSONNEL:

1. Doors to all laboratories should be locked on leaving the premises in the


evening and at weekends.
2. All lights and items of electrical equipment should be turned off (unless
otherwise indicated).
3. The last person to leave should also check that everything is in order before
going home and ensure that the premises are properly secured.
4. Access to departmental laboratories before 9-00 am and after 5-00 p.m. each
day and at weekends shall be restricted to authorized personnel only.
5. If you see a device with a frayed wire or broken plug, immediately report it to
the instructor.
6. Never lift a heavy object by yourself. Always get other to help.
7. When lifting keep your back straight and only lift with your legs.
8. Never place heavy objects on shelves or stacks above your head.

Students of all the F.Y, S.Y, T.Y and final year should follow the following rules
strictly:

1. Students should reach respective practical lab at least 5 minutes before scheduled
practical time.
2. Students should enter the laboratory after putting up their apron.
3. Students should come ready with all requirements for particular practical.
4. Students should listen all the instructions and practical to perform in depth and
should ask for any of the difficulty to concerned teacher.
5. Students should handle all inflammable solvents with care and should never take
them near flame.
6. Students should handle acids and alkalis in fuming cupboard only.
7. Students should keep all the reagent and solvents at proper place after using them.
8. Students should take care that while performing experiments they do not cause
any harm to property of college.
9. Students should not use mobile phone inside laboratory and should work in
discipline.
10. Students should report both positive as well as negative results to concerned
teacher and get it signed on that particular day only.
11. After due permission from concerned teacher student should leave practical lab
silently.
12. Students should remove their apron after leaving respective practical laboratory.

GUIDE LINES FOR PERSONAL SAFETY:

1. Wear a white protective coat when enter in to a laboratory.

2. Good quality of disposable plastic gloves must be used profusely while handling
both corrosive and poisonous chemicals.

3. Use close fitting safety spectacle or vision covering the entire face for protection
in the event of chemical splashing or spraying of corrosive or toxic hot liquid or
gases.

4. Keep organic solvents away from burner.

5. Always use Steam bath/Hot plate for evaporating organic solvents.

6. Do not suck any Reagent/Solvent/Base/Acid etc.

7. All experiments involving toxic solvents and reagent should be carried out in an
efficient fume-cupboard.

GUIDE LINES FOR ACCIDENT OR INJURY: -

1. Use first aid kit-box (with Lab assistant) in minor injury.

2. Water should never be employed to extinguish fire produced from organic solvents.

3. Any type of fire extinguisher must not be used on person.

4. Ordinary bucket of sand is invariably used against fire.

5. In the event of an accident/injury resulting in a cut/wound immediately encourage it to


bleed, wash thoroughly with soap and water but DO NOT SCRUB, then cover with a
waterproof dressing. In event of contamination of skin, conjunctivae or mucous
membrane immediately wash affected area thoroughly with sterile clean water.

6. Fire. Personal safety is most important. If a person’s clothing catches on fire, he/she
needs help. Prevent him/her from running. If he/she is close enough, put him/her under
safety.

Dissolve 1.62gm anhydrous ammonium vanadate in 125 ml conc.
Sulphuric acid. Coo it and add 125ml of ice-cold water.
 1 N KOH
Dissolve 56.11gm Potassium Hydroxide in 100 ml Dist. Water.

 10% Ammonia
Take 10ml Conc. Ammonia + 90ml Water.

 10% KMNO4
Dissolve 10gm KMNO4 in 100ml water.

 10% NaOH
Dissolve 10gm NaOH in 100ml water.

 10.5% FeCl3
Dissolve 10.5gm FeCl3 in 100ml water.

 25% Alcoholic HCl


Take 25ml Conc. HCl + 75ml Alcohol.

 5% Na2CO3
Dissolve 5gm Na2CO3 in 100ml water.

 Alcoholic Picric Acid


Dissolve 5gm Picric acid in 100ml Alcohol.

 Bacl2 Sol.
Dissolve 10gm Bacl2 in 100ml Water.

 CaCl2 Sol
Dissolve 10gm Cacl2 in 100ml Water.

 Copper Acetate
Dissolve 10gm Copper Acetate in 100ml Water.

Mix 0.5gm of Copper Carbonate in water with 10ml distiller water and
gradually add to it 10ml of strong solution of Ammonia with constant stirring.

 Dilute H2SO4
Take 10ml Conc. H2SO4 + 90ml Water.

 Dilute HCl
Take 10ml Conc. HCl + 90ml Water.

 Dragandroff Reagent
Boil 14gm of Sodium Iodide with 5.2 gm basic Bismuth Carbonate in 50ml
glacial acetic acid for a few minutes. Allow it to stand overnight and filter off the
precipitate of Sodium Acetate crystals. To 40ml of the red brown filtrate add 160ml
of glacial acetic acid and 1ml water. Store it in bottle. When needed add 20ml of
Acetic acid to 10ml of stock solution and add up to 100ml water.

 Fehling Solution A
Dissolve 34.66gm of Copper Sulphate in distilled water and make volume
up to 500ml.

 Fehling Solution B
Dissolve 173gm of Potassium Sodium Tartarate and 50gm of Sodium
Hydroxide in distilled water and make volume up to 500ml.

 Hagers reagent
A saturated solution of picric acid is used for detection of alkaloids.

 Iodine Sol.
Dissolve 2gm Iodine and 3gm Potassium Iodide in 100ml water.

 Lead Acetate Sol.


Dissolve 50gm Lead Acetate in 500ml water. Warm the solution and kept
it overnight. Filter the solution and store in a bottle.

 Mayer’s reagent
Dissolve 1.Mercuric chloride in 60ml distilled water. (a)

Mix (a) and (b) and adjust the volume to 100ml with distilled water.

 Molisch Reagent
Take 10gm alpha Napthol and dissolve it in 100ml 95% alcohol.

Take 2gm Phloroglucinol and dissolve it in 10ml water and 90ml Alcohol.

 Picric Acid
Dissolve 5gm Picric Acid in 100ml Water.

 Ruthyenium Red
Dissolve 0.008gm Ruthenium red in 10ml of 10% lead acetate solution.

 Safranin
Make 1% solution of safranin in 50% alcohol.

 Sudan Red
Dissolve 0.04gm of Sudan Red in 20ml of 90% alcohol and 20ml glycerin.

 Vanilin-sulphuric acid
It is a freshly prepared solution of 0.5% vanillin in sulphuric acid-ethanol.

Dissolve 1.27gm of Iodine and 2gm of potassium iodide in 5ml of water and

Equipment: Compound Microscope

Procedure: -
1. Place the microscope on a stable place on a bench of suitable height free from
vibrations.

2. Provide illumination from any of the three sources, i.e., built-in lamp, external
lamp or sunlight.

3. Direct the path of light to pass through diaphragm, condenser, and the hole of the
stage with maximum intensity while setting the mirror. Use the concave side of
the mirror if there is no condenser and use the planeside if there is a condenser;
keep the condenser at the top position.

4. Place the slide-bearing object to be examined, between the clips on the


mechanical stage.

5. Revolve the nosepiece and align the low power objective 10X to examine the
object on the slide [objective must click into place].

6. Look through the eyepiece and not into it. Adjust the illumination to improve the
contrast. In case of low power, the illumination is cut down to minimum by
reducing the aperture size.

7. Put one hand on the focusing knob [coarse or fine as the case may be], and other

on the screw to move the stage. Use the course adjustment knob for bringing the
object to the focus and get a sharp focus with the fine adjustment knob. The lower
power objective is commonly used to screen the field of the view. Bring the
object of interest in the centre.

8. Switch to high power [40X] and increase the illumination as needed. While
changing the objective by rotating the nose piece make sure that the objective
clicks into place. Repeat the process of focusing as described earlier by using the
coarse adjustment knob followed by fine adjustment knob. Normally modern
microscopes being parfocal, once the object is focused under any objective, it
stays in focus for all objectives and no further adjustments are necessary.

9. After screening under low power and examining under high power, oil-immersion
objective is used for obtaining greater details of the object.

10. Place a small drop of oil on the slide, over the path of the light. Increase the
illumination to maximum. Turn the nosepiece and set the oil immersion objective
in its place.

11. Use fine adjustment knob to get the object in focus. If this fails, use coarse
adjustment to focus the slide.

12. With the use of fine adjustment, bring the object sharply into focus. It is often
necessary at times to raise or lower the condenser so that the optimum
illumination is secured. This is termed ‘critical illumination’.

13. Finally when the observation is finished, the oil adhering the slide is removed by
means of xylene and wiping it clean by means of lens paper.

Routine Care of the Microscope


Microscopes are delicate and expensive optical instruments. A few simple
precautions will prevent damage or excessive deterioration.

1. Carry the microscope by holding its arm or limb with one hand and the other hand
under the foot. Never swing the microscope while carrying it nor turn it upside
down.

2. Never ram an objective into a slide. Never force anything.

3. Never touch any optical surface with anything other than lens tissue.

4. At the end of the experiment, gently clean all immersion oil off the lenses. Do not

use any organic solvents [eg. ethanol, ether, xylene or toluene] to remove the oil
on a regular basis. This may dissolve the cement holding the lens in the socket.

5. Dust and fungus are the worst enemies of microscope. Fungus growing on lenses
and scratches caused by dust particles on the lens surface can ruin the microscope.
Hence, when not in use store the microscope away from the dust and humid
environment.

6. When in doubt, ask your instructor for assistance.

1. Keep the water bath clean and replace the water frequently when it appears dirty.

Procedure: -

1. Plug the balance in A.C. mains.


2. On the toggle switches which consists of Scale lamp and Heating bulb
3. Rotate the Light control button in clockwise rotation, which is used to heat of
infrared bulb.
4. Put on the infrared bulb for say 5 – 6 mins so as to evaporate the moisture on the
pan and then turn off infrared bulb.
5. Then rotate the right hand knob (A) and bring the needle (pointer) and scale to
coincide 100. If needle is not projecting 100, rotate left hand knob (B) to bring the
pointer at 100.
6. Lift the window of oven and bring the scale to position ‘0’ by rotating right hand
knob. Add sample (about 3 to 4 gm) to be tested in the pan so that the pointer
coincide to zero.
7. Now shut the window and ON the bulb again. Let it heat to 1200C, approximately,
for 10 – 15 min. The needle will lift upwards. Again rotate the Right hand knob
(A) to bring the needle and scale to coincide.
8. The reading on scale with pointer in its position shows percentage of moisture
content in the sample.

Precaution:

1. The balance should be placed on heavy table approximately leveled.


2. Remove the sample after taking reading and clean the pan.
3. Keep the temperature in control so that the sample may not be burned or damaged.
