The inflammatory response is characterized by accumulation of neutrophil cells around the

inflammatory tissue. TNF-α is a proinflammatory mediator stimulated by neutrophils which is
useful for stimulating inflammatory cells. The plants that are often used as wound healers are
binahong [Anredera cordifolia (Ten.) Steenis], the most widely used part of the treatment from
this plant is the leaves. This study aimed to assess the topical effectiveness of ethyl acetate
fraction of Binahong ( Anredra cordifolia (Tenore) Steenis) leaves on TNF-α levels and the
number of neutrophils in the new open wound in white rats Sprague-dawley strain. Research is
an experimental laboratory test in vivo, with the design of the Pre and Post Test Only Control
Group Design. Research was conducted in January to July 2019 at the Biomolecular and Animal
House Laboratory of the Faculty of Medicine, Palembang Sriwijaya University.The subjects of
the study were male white rats Sprague-Dawley strain that met the inclusion criteria. Total
subjects are 30 individuals divided into 5 groups, namely the negative control group (ointment
base), positive control group (povidon iodine ointment 10%), group Ethyl Acetate Fraction of
Binahong Leaf concentration 1%, group Ethyl Acetate Fraction of Binahong Leaf concentration
of 2% and group Ethyl Acetate Fraction Binahong Leaf concentration 4. The data obtained were
then analyzed statistically using the program SPSS version 21.0 with Saphiro-Wilk Test, Levene
Test, Mann Whitney test paired t test (Paired t-test), Unpaired t test (Independent t-test),
confirmation of Post Hoc using LSD. Histopathological examination results of the number of
neutrophils of rat skin tissue on day 3 group Ethyl Acetate Fraction of Binahong Leaf
concentration of 1%, 2% and 4% concentrations were as effective as Povidon Iodine 10% for
increasing neutrophil amounts. However there was the lowest increasing in neutrophils in group
Ethyl Acetate Fraction of Binahong Leaf concentration of 4%, at TNFα levels rat skin tissue on
the third day using ELISA reader, in group Ethyl Acetate Fraction of Binahong Leaf
concentration of 1% there was a significant decrease in TNFα levels compared to Povidon Iodine
10%.
