I. INTRODUCTION:
There are four broad classes of biomolecules that can be found in living systems. Each type of biomolecule
has a characteristic structure and function in living organisms. You can use your knowledge of the basic
structure of each biomolecule to perform tests in the laboratory that detect the presence or absence of key
functional groups or overall characteristics in various substances. In this experiment, you will utilize the
following procedures to detect the biomolecules present in a given substance.
II. OBJECTIVES:
1. To use indicators to test for the presence of lipids, carbohydrates and proteins in various foods.
2. To identify the type of biomolecule pesent in a type of food
III. MATERIALS:
Iodine solution Brown bag
Sodiul hydroxide Test tubes
Copper sulfate solution Beaker
Benedict’s solution Well plate
Food samples Test tube holder
IV. PROCEDURES:
TEST 1: Testing for Lipids
1. Tear off a piece of brown paper bag 30 cm long.
2. Draw six small squares, approximately 3 cm each, and label each with the name of the food samples.
3. Put 1 drop of each of the food samples in corresponding boxes on the brown paper.
4. Put the brown paper aside while you do the other tests.
5. When the brown paper is dry, record your observations in your data table. Look for a grease stain.

TEST 4: Testing for Proteins
1. Fill six wells in your well plate with food samples.
2. Add 4-5 drops of sodium hydroxide solution to each food sample.
3. Then add 2-3 drops of copper sulfate solution to each food sample.
4. Check for any color change and record data in your data table.
5. Clean and dry well plate.
