BIOMOLECULE TEST

I. INTRODUCTION:
There are four broad classes of biomolecules that can be found in living systems. Each type of biomolecule
has a characteristic structure and function in living organisms. You can use your knowledge of the basic
structure of each biomolecule to perform tests in the laboratory that detect the presence or absence of key
functional groups or overall characteristics in various substances. In this experiment, you will utilize the
following procedures to detect the biomolecules present in a given substance.

II. OBJECTIVES:
1. To use indicators to test for the presence of lipids, carbohydrates and proteins in various foods.
2. To identify the type of biomolecule pesent in a type of food

III. MATERIALS:
Iodine solution Brown bag
Sodiul hydroxide Test tubes
Copper sulfate solution Beaker
Benedict’s solution Well plate
Food samples Test tube holder

IV. PROCEDURES:
TEST 1: Testing for Lipids
1. Tear off a piece of brown paper bag 30 cm long.
2. Draw six small squares, approximately 3 cm each, and label each with the name of the food samples.
3. Put 1 drop of each of the food samples in corresponding boxes on the brown paper.
4. Put the brown paper aside while you do the other tests.
5. When the brown paper is dry, record your observations in your data table. Look for a grease stain.

TEST 2: Testing for Starches


1. Fill the spaces in your well plate with the food samples.
2. Add 10 drops of iodine solution to each food sample.
3. Observe and check for any color change then record data in your data table.
4. Clean and dry well plate.

TEST 3: Testing for Sugar


1. Put one dropper full of each food samples in six different test tubes. Make sure to labe all test tubes.
2. Add 10 drops of Benedict’s solution to each test tube and place them all carefully into the hot water bath
for 3-5 minutes.
3. Remove test tubes from hot water bath using designated tongs or test tube holder and place them into a
test tube rack. Observe for the change in color and record it in your data table.
4. Wash all test tubes and place in test dube racks upside down to dry.

TEST 4: Testing for Proteins
1. Fill six wells in your well plate with food samples.
2. Add 4-5 drops of sodium hydroxide solution to each food sample.
3. Then add 2-3 drops of copper sulfate solution to each food sample.
4. Check for any color change and record data in your data table.
5. Clean and dry well plate.
