The effect of different concentrations of sucrose on the size of potato discs was determined
by carrying out the following steps:

Step I: Using a cork borer, 5 cylinders of potato tissue were obtained. Each cylinder was then
sliced to make 10 discs of uniform size.

Step 2: 10 discs were taken and measured as shown in Diagram 1.1

Step 3: The discs were immersed in 100 ml of distilled water in a Petri dish labelled J.
Step 4: Steps 2 and 3 were repeated for the remaining discs which were then placed in Petri
dishes labelled K,L,M and N, each containing sucrose solutions as shown in
Table 1.1.

The rate of enzyme reaction is influenced by temperature. When the temperature is low,

the enzyme is not active. When the temperature increases, the enzymes are more active.
Enzymes worked best at the optimum temperature and become denatured if the
temperature is increased above the optimum temperature.

Kadar tindak balas enzim dipengaruhi oleh kesan suhu.Apabila suhu rendah, enzyme
tidak aktif. Apabila suhu meningkat, enzim semakin aktif. Enzim sangat aktif pada suhu
optimum dan mengalami denaturasi apabila suhu melebihi suhu optimum.

Based on this statement, design an experiment to show the effect of temperature on the rate
of reaction on the enzyme amylase on starch.
