In Vitro testing of the antifungal properties of Cyperus Rotondus (Nutgrass) against Candida Albicans

RATIONALE

Candida Albicans is a kind of yeast that causes the disease


Candidiasis which affects the vagina of women and sometimes the throat
(Schulman, 2018). The vaccine of the Candidiasis is the Flucanozole, which have
some side effects like headache (Pagano. 2019.) According to Rxlist, the
component of the Flucanozole that cures the Candidiasis is the Povidone. Povidone
is a type of Iodine which can be going on plants like nutgrass when planted on soil
which is Rich in Iodine (Forberg & Myhre, 2017).
Candida Infections are prevalent, debilitating and frequently recurrent
fungal diseases and a clinically important issue. Candida Albicans is the Candida
spp's most virulent. Can cause serious mucosal and life threatening systemic
diseases in hosts with immunocompromise. We chose nutgrass for this reason. It is
astringent, diaphoretic, diuretic, analgesic, antipasmodic, aromatic, carminative,
antitussive, emmenagogue, litholytic, sedative, stimulant, stomachic, vermifuge,
tonic, and antibacterial rhicomes are regarding to Ayurveda ( n.d. ).
It can be a nice indigestion remedy. The scientists have decided to
conduct the in vitro testing in the Capitol Medical Center. The scientists studied its
background and found out that they can conduct in vitro experiments. In practice
the development of in-vitro tests most likely will proceed in at least 2 distinct
phases: identification by a series of individual tests of the fundamental biological
properties of the test drug; and findings detected in the phase 1 tests would be
investigated in more sophisticated versions of the test, and these phase 2 tests
usually would be of much longer duration. Table 1 lists the vast range of cells and
tissues which have been used for in-vitro tests and details some of those most
relevant to preclinical testing of drugs in the field of fertility regulation. The tests
fall into the categories of microbiological tests, fungal cultures, tissue culture
teratogen assays, cytoxicity tests -- sensitivity of tissues, and sperm tests in
toxicology. (Pearson RM., 1986)
In Vitro Method: Agar disk-diffusion method
In this well-known procedure, agar plates are inoculated with a
standardized inoculum of the test microorganism. Then, filter paper discs (about 6
mm in diameter), containing the test compound at a desired concentration, are
placed on the agar surface. The Petri dishes are incubated under suitable
conditions. Generally, antimicrobial agent diffuses into the agar and inhibits
germination and growth of the test microorganism and then the diameters of
inhibition growth zones are measured.

The society will benefit especially for those who have the fungal
infections called Candidiasis. The benefit of this is it can treat Candidiasis using
Nutgrass which is easily found. The significance/importance of this is to lessen the
case of the fungal infection called Candidiasis and also to know more of what the
Candidadiasis is, and for us to become aware of its symptoms and causes.

STATEMENT OF THE PROBLEM


This research aims to answer the effectivity of the Antifungal property of
Nut grass against Candida Albicans in terms of:
1. Will Cyperus Rotondus (Nutgrass) extract be able to exhibit fungicidal
effects against Candida Albicans in terms of inhibitory activity?
HYPOTHESIS
The Nutgrass will show no signs of antifungal property to Candida Albicans
in terms of the Zones of Inhibition.
Goals
The scientist’ goals are to prove that Cyperus Rotundus (Nutgrass) have the
antifungal properties against Candida Albicans using In Vitro testing to lessen the
cases of Candidadiasis.
Expected Outcome
The scientists’ expect to lessen the case of Candidadiasis, Athlete’s Foot,
Diaper Rash and Jocks’ Foot because the diseases are one of the most common
diseases caused by Candida Albicans

Collection of Materials

The scientists will buy approximately 1kilogram of Nutgrass from the Karuhatan
Wet Market or the palengke in Barangay Parada. The scientists will also buy a
strain of Candida Albicans at the National Institute of Microbiology and
Biotechnology in the University of the Philippines, Los Baños.

Preparation

The nutgrass will be grinded by a mortar and pestle and the Candida Albicans will
be placed in a petri dish.
