DISCUSSION

The homogenizing solution is important in the experiment, it allows the release of the DNA from
the clinging proteins. Homogenizing solution served as the control test in the experiment. The solution is
made up with hot distilled water added with 1.5 NaCl and a 5 ml dishwashing liquid. It was stirred
cautiously to avoid foaming.

In the extraction of DNA, the banana was threaded then was added a hot distilled water during
homogenization. The thermal treatment was performed to relieve the cell and separate the cells. The
phospholipids of the membrane were damaged by the hot water and the DNase denatured, as if this
were present, the DNA would fragment. A thread-like structure was created in the ethanol layer after
homogenizing the sample and placing it into cold ethanol. DNA isolate was the surface. Furthermore,
DNA is soluble and invisible in water, however, it is insoluble in alcohol in which it precipitates and
becomes visible. In addition, the ethanol removes other impurities in the banana filtrate and precipitate
pure DNA. The banana tissue is mixed in a blender with the homogenizing solution, which breaks down
the cell wall, cell membrane and nuclear membrane allowing the release of DNA. It was then spooned
and was added SSC solution.
