Lab Report

Experiment no. 1
Title: pH And Buffers
Biochem Lab, BSN 1B Date Submitted:
Group no. 3 Date Performed:
Class Schedule: Friday, 7am – 1pm
Group Members:

I. Objectives
1. To illustrate the buffering properties of phosphates and acetates.
2. To explain how buffers work.
3. To know the effects of acid to both buffers and weak base (H20).

II. Introduction:
The term pH was introduced in 1909 by Sorensen, who defined pH as negative log of hydrogen
ion concentration. The pH is a figure expressing the acidity and alkalinity of a solution on a logarithmic
scale on which 7 is neutral, lower values are more acid and higher values are more alkaline. According to
Bronsted-Lowry definition, acids are proton donor and bases are proton acceptor. A buffer is a solution
containing either weak acid or a weak base which is resistant to changes in pH. Buffers are also used to
maintain stable pH in a solution, as they can neutralize small quantities of additional acid of base.
III. Materials:

IV. Procedure (Schematic Diagram):

V. Results:

MIXTURE pH Meter pH Paper

KH2PO4 and K2HPO4 6.5 7 -Neutral
CH3COOH and CH3COONa 4.9 4- Acid
Distilled Water 7 7- Neutral
Distilled Water + HCl 3.7 5 -Acid
Phosphate Buffer + HCl 6.5 7 -Neutral
Acetate Buffer + HCl 4.6 4 -Acid

VI. Discussion:

VII. Conclusion:
As for the conclusion, Buffer solutions are solutions that resist change in Hydronium ion and the
Hydroxide ion concentration (consequently pH) upon addition of small amounts of either acid or base, or
upon dilution. Buffer is mixture of a weak acid and its conjugate base. Because the solute can either
absorb protons or release protons, buffer solution can maintain the pH at a constant value.
From the results obtained, it can be concluded that the initial pH value for the Phosphate buffer was 6.5
and when HCl acid was mixed the pH value of Phosphate buffer plus the HCl acid remains the same and
the level of acidity w
