Laboratory Report

Pre-lab Questions:
1. What is the function of chloroplasts?

2. Name two structures found in plant cells but not in animal cells.

3. Name three structures found in plant and animal cells.

4. What structure surrounds the cell membrane (in plants) and gives the cell support.

5. What are the structures that you may found on bacteria cell.

Laboratory:
Part 1: Onion Cells
Method:
1. Obtain a thin slice of onion
2. Put a drop of iodine stain on slide. (Be careful stains can ruin clothing)
3. Place onion slice in iodine stain on the slide. The slice should not be wrinkled or folded.
4. Put a cover slip over the stain.
5. Observe under the microscope under low, medium, and high power.
6. Draw your observations on your paper for low, medium, and high power.
a. Label the cell membrane, nucleus, cytoplasm, and any other organelles you observe (i.e.
vacuoles)
7. Place your used materials in the designated area.

Part 2: Epithelial or cheek cells


Method:
1. Obtain a clean slide and cover slip.
2. Put a drop of iodine stain on a slide.
3. Gently rub the end of a toothpick on the inside of your mouth along the cheek. (Be careful. Do
not gouge out your cheek.)
4. Stir the toothpick in the iodine stain
5. Immediately, throw the toothpick away.
6. Put a cover slip over the stain.
7. Observe under the microscope under low, medium, and high power.
8. Draw your observations on your paper for low, medium, and high power.
a. Label the cell membrane, nucleus, cytoplasm, and any other organelles that you observe.
Part 3: Yakult/Yogurt Cells
Method:
1. Take a very small drop of yogurt with the toothpick or a small drop of yakult and smear it for
2-3 seconds on the slide.
2. Put a drop of Iodine stain on the slide. (Optional)
3. Put a cover slip over the yakult/yogurt
4. Observe under the microscope under low, medium, and high power.
5. Draw your observations on your paper for low, medium, and high power.
a. Label the cell membrane, cell, wall, cytoplasm, and other organelles you observe.

Post-Laboratory:
1. Describe the shape and the location of chloroplasts.

2. Why were no chloroplasts found in the onion cells?

3. Make venn diagram to show the differences and similarities between the onion cells and
epithelial/ cheek cells.

4. Reflection: Share your experience before, during, and after the experiments.
