This Commissioning Test Plan (CTP) is for the AHU-101 air Handling Unit in 1 st Interstitial Floor for the Reliance
Life Sciences Pvt Ltd. (RLS) cGMP Plant2 project at Navi Mumbai, India.

2. Purpose

This commissioning test plan describes the testing/verification activities that will be completed/documented to
verify that the system is ready for turnover to qualification (for direct impact systems) or to the system owner
(indirect and no impact systems). For direct impact systems, commissioning will further allow for the resolution
of punch list items prior to carryover as deviations to the qualification phase.

The purpose of commissioning is to provide a well-planned, documented approach to ensure that start-up and
turnover of each system results in an operational, safe, and functional system that meets user requirements and
operational expectations. This commissioning document will be executed by completing a series of data
sheets.

Commissioning activities are divided into three phases:

 Inspection
 Regulation, Adjustment, and Setting to Work
 Functional Testing

6 System Functional Testing

6.1 AHU Operational Test

Test Procedure:
 Verify all the equipments, Gauges are calibrated and factory tested.
 Verify all Installation checks are completed, punch list attended and resolved.
 Check all safety checks done.
 Check on smooth, quite and vibration free operation of Fan motor
 Check current, rpm for Fan and motor
 Record Air quantity and flow rate of Chilled and Hot water.
 If readings are more/ less compared to Design parameter balance the air and water sides using
control devices like Dampers, Balancing Valves etc.


6.2 VFD Operation Testing

This test verify the VFD and static pressure sensor (PT/PE) is operating as specified and
controlling the speed of supply fan (PY) to meet desired discharge static pressure.

6.3 HEPA Filter Testing

6.3.1. Air Velocity and Uniformity Test.

These test procedures are performed to determine average airflow velocity and uniformity within a clean
room, clean zone or unidirectional flow work zone, as well as to determine air supply volume flow rate
and volume uniformity.

Pre-Requisites / Reference Documentation

 Air flow velocity: electronic micro manometer with tube array, thermal anemometer, vane
type anemometer or equivalent.
 Air flow volume: electronic macro manometer with appropriate airflow hood or equivalent.
 Suitable support stands where necessary.

Pre-test criteria:
 The filter should be completely installed and sealed properly.
 AHU system should be in running condition
 Air flow through the filter to be adjusted and balanced.

Testing Procedure

This test is performed to determine the average airflow velocity uniformity range within unidirectional/non
unidirectional clean rooms.

 Divide the effective HEPA filter face area into equal grids not greater than one square
foot starting six inches in from the inner edge of the filter frame.
 Measurement should be taken at the center of each grid cell.
 Measuring time at each position should be at least 5 seconds duration.
 The minimum, maximum and average values should be recorded.
 Air Volume is then calculated (CFM) by multiplying the effective filter area by the average
velocity

(Effective filter area) ft2 x (Average filter velocity) FPM = (Volume) (CFM)
Note: Air changes can be calculated by Velocity (FPM) x filter area (FT 2) x 60 divided
room volume.

 The average airflow velocity, or the average total airflow volume, for the clean room or
clean zone should be within ±5% of the value specified for the clean room or clean zone,
or within other tolerance limits agreed upon by the buyer and the seller.
 For Class 100 areas, airflow rate of minimum of 240 air changes per hour.
 For Class 10,000 areas, airflow rate of minimum of 50 air changes per hour.
 For Class 100,000 areas, airflow rate of minimum of 25 air changes per hour.

6.3.2. HEPA filter Integrity Test

To confirm that The HEPA Filter System is properly installed by verifying the absence of bypass leakage
in installation and that filters are free from small leaks and defects.

Pre-Requisites / Reference Documentation

 Aerosol Photometer duly calibrated.
 DOP Generator.
Leak Test Procedure
Pre-test criteria:
 The filter should be completely installed and sealed properly.
 AHU system should be in running condition
 Air flow through the filter to be adjusted and balanced.
Testing Procedure
This test is performed by introducing the specified challenge aerosol upstream of the filters and
searching for leaks by scanning the downstream side of the filters with the photometer probe.
The design airflow velocity should be established and uniformity confirmed according to section 1
prior to performing this test.
The air-generated aerosol challenge and aerosol photometer filter scan test should be performed
as follows:

 The filter must be completely installed and flow uniformity adjustment to be completed.
 An aerosol generated by blowing the compressed air through Di Octyl Phthalate
generator.
 Inject the aerosol at mixing chamber of the air handling unit.
 Place the aerosol generator in an accessible position, and adjust the aerosol
concentration between 10 to 20 microgram per liter. The set compressed air supply
pressure to DOP generator should be 44-PSI minimum.
 The photometer has the sample flow rate of 1 cfm, The probe is held at a distance of 6
inch and at a traverse rate of 3m/ min
 The filter face and perimeter is scanned by passing the probe covering the complete
area so that the entire area of the filter and sealing gasket is sampled
 Scan the filter thoroughly to determine the leakage produced due to DOP sensing during
the test.

6.3.3. Room Pressurization Test

The purpose of this test is to verify the capability of the cleanroom system to maintain the pressure
differential between the cleanroom and its surroundings.

Pre-Requisites / Reference Documentation

 This test should be performed after the facility has met the acceptance criteria for air flow
velocity or volume, airflow uniformity, parallelism and other applicable tests.
 Apparatus for the room pressurization test should consist of an electronic micro-
manometer, inclined manometer or mechanical pressure gauge.

Testing Procedure
With all doors closed, the procedure should be performed as follows:

 Measure and record the pressure differential between the clean room an anteroom (if
present), and between the anteroom and the external environment.
 If no anteroom is present, measure and record the pressure differential between the
cleanroom and the external environment.
 If the clean space is subdivided into more than one room, measure the pressure
differential between the innermost room and the next room in order. Continue until the
last room (or anteroom) has been measured against the external environment.

6.3.4. Airflow Parallelism Test

The purpose of this test is to verify the parallelism of airflow throughout the work zone and whether the
cleanroom is capable of limiting the dispersion of internally generated contamination.

Pre-Requisites / Reference Documentation

 This test is applicable for unidirectional air flow only.
 This test should be performed after the facility has met the acceptance criteria for air flow
velocity or volume, airflow uniformity and other applicable tests.
 An isokinetic smoke generator capable of supplying a smoke source at a specified
discharge speed is required.

Testing Procedure
 Divide the entrance and exit planes of the work zone into grids of equal area, having
dimensions of 3m x 3m (10 ft x 10 ft).The minimum distance between entrance and exit
planes should be 0.9m (3 ft).
 Set up the smoke generator with the outlet tube pointing in the direction of flow and
located at the center of the grid area at the work zone entrance plane. Smoke generation
should be performed outside of this grid area.
 Introduce smoke isokinetically and isothermally.
 Measure the offset distance between the theoretical straight-line flow point and the radial
direction of offset.

Acceptance Criteria and Test Results

 Readings must not exceed angle of 14 degrees.

6.3.5. Enclosure Induction Leak Test

This test is performed to determine if there is intrusion of unfiltered air into the clean work areas from
outside the cleanroom enclosure through joints, seams, and doorways.

Pre-Requisites / Reference Documentation

 Air – generated aerosol source or acceptable substitute.
 Discrete – particle counter having a particle size discrimination capability of 0.5 µm or
smaller.
 An isokinetic smoke generator capable of supplying a smoke source at a specified
discharge speed is required.

Testing Procedure
 Measure the particle concentration outside the cleanroom enclosure immediately
adjacent to the surface or doorway to be evaluated. This concentration should be at least
3.5 x 10 particles /m³. (If the concentration is less, generate an aerosol to increase the
concentration.
 To check for leakage through construction joints or cracks, scan at a distance of 5 to
10 cm (2to 4 inches) from the joint at a rate of approximately 3 m / min (10ft / min.).
 To check for intrusion at open doorways, measure the concentration inside the enclosure
at 0.3 to 3 m ( 1 to 10 ft) from the open door.

concentration.

6.3.6. Temperature Uniformity and Humidity Test

The purpose of this testing s to demonstrate the capability of the cleanroom air handling system to
maintain temperature and humidity level within the control limits over specified period of time.

Pre-Requisites / Reference Documentation

 Psychometer and temperature/humidity sensor.
 Ensure that the accuracy of the sensor is register.
 Each zone allowed operating minimum of 24 hours before testing is begun.

Testing Procedure
 Divide the area into equal grids approximately 10ft x 10ft .
 With one indicator at work height the center of one grid, temperature is measured in all
the other grids with the second sensor, each time reading both meters simultaneously.
 Two temperatures are recorded at each data point.

Acceptance Criteria and Test Results

 Temperature and humidity is throughout the area within the following range 22 o C ± 2 o C
 RH should be 40 to 50 %.
