ACTIVITY NO. 1
MICROSCOPE MANIPULATION

Objectives:
This activity aims to give an opportunity to familiarize with the essential and basic
concepts in using a microscope.
Materials:
 Microscope
 Glass slides
 Letter “e” from a newspaper
Procedure:
 Place the microscope on the table and set up for the activity
 Once the microscope has been set, prepare the specimens to be used
 On a glass slide, place each specimen carefully and lay them out flat
 Position the mirror so that there is enough amount of light to see the specimen
 Start with the low-power objective lens and adjust until the specimen is identified
 Move the adjustment knob to adjust focus
 Carefully switch objective lens from the lowest power to highest power and observe
 Record the result

Discussion:
Comparing the three different lenses, the scanning objective lens was quite easy to
use. Rotating the adjustment knob and moving the specimen in the stage helps in finding the
specimen. After shifting to the next objective with low power, the vision become quite blurry
so all that was needed was to move the adjustment knob until the specimen was clearer to
see again. Based on my observation, using the low power objective enables the specimen’s
overall structure. But it still need more magnification to see clearly and dig deep into its parts.
Lastly, in using the high power objective lens, it was quite difficult because it needed a lot of
light that the mirror needs to reflect, patience and concentration. It was difficult to see the
specimen because it needs a lot of adjustment. Rotating the fine adjustment knob was able to
let me see the specimen more clearly. I observed that increasing the magnification allows me
to see the very small details and contents of the specimen.

Generalization:
The microscope is a very important tool to science. It allows a person to see the tiniest
things that our naked eye cannot. It takes a lot of patience, concentration and practice to be
able to use the microscope more efficiently. It was quite difficult at first, but as you familiarize
each part and uses of the microscope, the result are worth it. We are able to see the hidden
parts of everyday objects we commonly see.
Throughout the activity, I learned the basic and essential skills in using a microscope.
From setting, handling and manipulating the very microscope in order to see the magnified
view of the given specimen.
