The detergent salt solution is used to break down the fat and proteins that make up the cell
membrane. The sale causs the phosphate ends of the DNA to come closer together which will
make it easier to precipitate out of solution.
Alcohol is used to precipitate the DNA. Because DNA is soluble in water, alcohol (ethanol)
causes the DNA to precipitate and come out of the solution. DNA will rise into the alcohol layer.

Why use liquid detergent and salt?
Liquid detergent causes cell membranes to break down by emulsifying the lipids and proteins of the cell and disrupting the bonds
that hold the cell membrane together. The detergent causes the lipids and proteins to precipitate out of the solution. Salt enables
DNA to precipitate out of an alcohol solution because it shields the negative phosphate end of DNA, causing the DNA strands to
come closer together.
Why use warm water bath?
The heat treatment of water bath softens the phospholipids in the cell membrane and denatures the DNAse enzymes that, if
present, would cut DNA into small fragments so it cut not be extracted.
Why mash onion?
Mashing the onion helps break open the cells (cell walls, cell membranes and nuclear membrane, allowing the release of the
DNA).
Why use chilled ethanol?
DNA is soluble in dish detergent solution but insoluble if ethanol is added. Adding ethanol precipitates the DNA so you can see
it. Ethanol causes every component in the filtrate to stay in solution except the DNA.
