Methods 8001 and 8001A meet or exceed the specification criteria stated in Standard Methods for the Examination of Water and Wastewater, 19th edition, 9221 Multiple-Tube Fermentation Technique for Members of the Coliform Group. For potable water confirm fecal coliforms with EC Medium Broth as cited in 40 CFR Part 141.21, Subpart (F)(5); or confirm E. coli with EC/MUG Medium Broth as cited in 40 CFR Part 141.21, Subpart (F)(6)(i). ** Most Probable Number Method 8368, A-1 Medium, is USEPA-accepted for testing non-potable waters. Method 8368 meets or exceeds the specification criteria stated in Standard Methods for the Examination of Water and Wastewater, 18th edition, 9221 E. Fecal Coliform Procedure. USEPA Manual for the Certification of Laboratories Analyzing Drinking Water states 5.5.3. A-1 medium may be used as an alternative to EC Medium to enumerate fecal coliforms in source water, in accordance with the Surface Water Treatment Rule. A-1 Medium must not be used for drinking water samples.

Introduction
The Most Probable Number (MPN) method (also referred to as the Multiple Tube Fermentation Technique) uses a specified number of test tubes to statistically predict the number of organisms present (based on the expected population of organisms in the sample). The MPN method is ideal for wastewater samples and nonpotable samples, because the analyst can accommodate highly turbid samples by diluting prior to analysis. No filtering is necessary. The MPN method uses screw-capped tubes containing sterile broth medium. Tubes may also contain an inverted inner vial (Durham tube) for gas collection. Simply add sample to a tube of prepared medium and incubate. If coliforms are present, gas is produced and is trapped in the inner vial. Fluorescence can indicate the presence of E. coli.

Presumptive Coliform Test Methods 8001 and 8001a

1. Wash your hands thoroughly with soap and water.

2. Prepare any necessary dilutions.

3. Open the sealed package of the Lauryl Tryptose (LT) Broth For potable water: Analyze without diluting. tubes. For potable water: For nonpotable water: Remove 5 or 10 tubes. Using sterile buffered (The 5-tube test is not a dilution water, prepare 3 decimal dilutions of the USEPA-accepted method.) Use a sterile sample. See Diluting Samples in Introduction to pipette to transfer 10 ml of sample to each tube. (See Bacteria. note a on page 1146.)
For nonpotable water: Remove 15 tubes. Remove the caps from the tubes one at a time. Use a sterile pipet to transfer 10 mL of the first dilution into 5 tubes. Use a sterile pipet to transfer 10 mL of the second dilution into 5 tubes. Use a sterile pipet to transfer 10 mL of the third dilution into the remaining 5 tubes. This completes a 15-tube dilution series (5 tubes for each of 3 dilutions).
Note: Be careful not to touch the open end of the tubes or the inside of the caps.

4. Replace the screw cap on each tube immediately after sample addition. Invert the tube a few times to thoroughly mix the sample with the nutrient medium. After the last inversion, make sure the tube is upright and the inner vial is full of liquid with no air bubbles. The tube must be kept upright for the rest of the procedure.

Confirm Positive Results

5. Place the LT Broth tubes in a Dri-Bath or other incubator at a temperature of 35 0.5 C. After 1 hour, invert the tubes to remove trapped air in the inner vials; then continue to incubate. See note b on page 1146.

6. After 24 2 hours, check the inner vials for gas bubbles. If the broth appears cloudy (indicating bacterial growth) and the inner vials contain gas bubbles, coliform bacteria are presumed to be present. If no gas is present, return the tubes to the incubator and examine again after a total of 48 3 hours. Formation of gas in any amount, with growth, constitutes a positive test. Absence of gas at the end of this period constitutes a negative test. If gas and growth appear before 24 hours have elapsed, inoculate confirmation medium without waiting the entire 48 hours.

7. All LT Broth tubes showing gas formation should be confirmed for total coliforms and fecal coliforms or E. coli.
Note: A confirmation test is necessary to eliminate falsepositive results that sometimes occur when only the presumptive test is used.

Confirmation of Fecal Coliforms and E. coli Methods 8001 and 8001a
The USEPA requires that samples testing positive for total coliform bacteria also be tested for fecal coliform or E. coli bacteria, using EC Medium Broth or EC/MUG Broth, respectively.

1. From each positive LT Broth tube, inoculate an EC Medium Broth tube or EC/MUG Broth tube. EC Medium Broth is used to test for fecal coliforms. EC/MUG Broth is used to test for E. coli. Inoculate the confirmation tubes using a sterile, disposable loop or a nichrome wire loop that is flame sterilized. Transfer one loop from the positive LT Broth tube to a confirmation tube, making sure not to touch the rim of either tube. See note c on page 1146.
Note: To sterilize an inoculating loop, heat to red hot in an alcohol or Bunsen burner. Let the loop cool before use.

Note: Until you gain confidence in differentiating between fluorescent and nonfluorescent samples, include a tube containing a known E. coli culture with each set of tests. Note: Fluorescence without gas production indicates an anaerogenic strain(s) of E. coli. Note: The USEPA requires that samples testing positive for total coliform bacteria should also be tested for fecal coliform or E. coli bacteria, using EC medium Broth or EC/MUG Broth, respectively.

Test for Total Coliforms and E. coli (LT/MUG) Method 8091*

Hachs LT/MUG (Lauryl Tryptose with MUG) Method** is a rapid system for simultaneous detection of total coliforms and E. coli. The results are comparable to the traditional MPN fecal coliform tests, but obtained in far less time. No transfer from presumptive to confirmed medium is necessary with the LT/MUG method. The LT/MUG medium contains lauryl tryptose broth and 4-methylumbelliferyl--D-glucuronide (MUG), a fluorogenic reagent. Tubes positive for E. coli will fluoresce when the incubated tubes are examined under long-wave UV light.

Absence of gas constitutes a negative test for total coliform bacteria. Tubes that contain growth, but without gas production, should be checked for fluorescence.

fluorescence by using a long-wave ultraviolet (UV) light. Make sure to examine tubes in a darkened area. Tubes that exhibit fluorescence are positive for E. coli. If the sample does not fluoresce, continue incubation for a total of 48 3 hours. Absence of fluorescence constitutes a negative test for E. coli.
Note: Until you gain confidence in differentiating between fluorescent and non-fluorescent samples, include a tube containing a known E. coli culture with each set of tests. Note: Fluorescence without gas production indicates an anaerogenic (non-gas-producing) strain(s) of E. coli. Note: Fluorescence confirms a positive E. coli test and additional tests for fecal coliforms should not be necessary. However, even though studies indicate fluorescence in LT/MUG Broth is superior to traditional method for confirming the presence of E. coli, all types of samples have not been tested under all circumstances. To ensure LT/MUG results are comparable to fecal coliform numbers acquired from traditional methods, you should run several parallel tests using both procedures.

Test for Fecal Coliforms (A-1 Medium Broth)

Method 8368

1. Wash hands thoroughly with soap and water.

2. Using sterile buffered dilution water, prepare 3 decimal dilutions of the sample. See Diluting Samples in Introduction to Bacteria.

3. Open the sealed package of the A-1 Medium Broth tubes and remove 15 tubes. Remove the caps from the tubes one at a time. Use a sterile pipet to transfer 10 mL of the first dilution into 5 tubes. Use a sterile pipet to transfer 10 mL of the second dilution into 5 tubes. Use a sterile pipet to transfer 10 mL of the third dilution into the remaining 5 tubes. This completes a 15-tube dilution series (5 tubes for each of 3 dilutions).(See note a on page 1146.)

6. At the end of
24 2 hours, tap each tube gently and examine for gas. If the inner vials contain gas bubbles, fecal coliform bacteria are present. When growth is present, formation of gas in any amount constitutes a positive test. Absence of gas at the end of this period constitutes a negative test.

Notes a. The presumptive tubes are specially designed to contain 10 mL of sample when the inner vial is empty (filled with air) and may be used to measure the amount of sample added when USEPA reporting is not required. When USEPA reporting is necessary, measure the sample by pipetting. b. It is important to check for air in the inner vials after 1 hour of incubation. Bubbles formed during this period result from dissolved gases released by heating and are not due to bacteria. When present, trapped air should be removed by inverting and carefully returning the tubes to their upright position. Loosen the caps slightly before returning the tubes to the incubator. c. Inoculation can be done by transferring the cap of the positive presumptive tube to the confirmation tube. The cap transfer method of inoculation is not acceptable for USEPA reporting. Shake or invert a positive presumptive tube to wet the liner of the cap. Exchange this cap with the cap of the confirmation tube and then invert the confirmation tube to mix the transferred bacteria with the nutrient medium.

Interpreting and Reporting Results
For potable water
The USEPA has established minimum requirements* for sampling, check sampling, and interpreting results. Certain areas may have more stringent requirements. If all 10 tubes (for a 10-tube MPN test) of the confirmed coliform test are negative, the sample is accepted as meeting bacterial standards. To assure that sample results are interpreted in accordance with appropriate standards and regulations, contact your local, county, state, or federal agency. Five-Tube MPN Test Use Table 1 to determine MPN/100 mL for 5-tube MPN tests. For example, if 4 of the 5 tubes showed a positive response, the MPN/100 mL would be 16.0.

Ten-Tube MPN Test Use Table 2 to determine MPN/100 mL for 10-tube MPN tests. For example, if 6 of the 10 tubes showed a positive response, the MPN/100 mL would be 9.2.

Using statistical methods, it is possible to estimate the number of coliform organisms from any combination of positive and negative test results. The MPN Table is based on three dilutions of decreasing decimal increments.

Examples

Five tubes of undiluted sample, five tubes of 10X dilution, and five tubes of 100X dilution are inoculated initially. Positive results are obtained from five of the undiluted samples, two of the 10X dilutions, and none of the 100X dilutions. The result is expressed as 5, 2, 0. In this example, the first number (5) in the coded result represents the use of an undiluted sample (dilution factor of 1), so the MPN per 100 mL is read and recorded directly from the appropriate column in Table without adjustment for dilution differences.

In Example B, the first number (5) in the coded result represents a sample dilution with a factor of 10 rather than 1, so Table on page 1148 cannot be used directly. The 5 of the 5, 1, 1 code in this dilution series actually represents the number of positive coliforms observed using a sample dilution of 1:10. Thus, a multiplication is required to give the corrected MPN. Use the tables as before, but multiply the MPN index of 50 by the smallest dilution factor used (in this case 10) to give the corrected value of 500. A simple formula for this type of correction is:
MPN Index from Table x smallest dilution factor used = MPN/100 mL.

Design the series of decimal dilutions so that it contains both positive and negative tubes. If all the tubes of the 3 dilutions are positive, a greater degree of dilution is needed. Similarly if all tubes of the 3 dilutions are negative, the degree of dilution is too great.

In this example, all tubes were positive. Thus, the dilution series was not high enough to yield a reliable MPN. The only information obtained indicates a count greater than 160,000 coliforms/100 mL.
