How to choose an SPE product


1. Characterize the sample
Factors such as the analytes polarity relative to the matrix, the presence of charged functional groups, solubility, molecular weight, etc., determine how strong the analyte is retained by the packed bed.

Process individual samples via luer hub syringe. Connect syringe directly to SPE cartridges or to SPE columns via adapters.

2. Select a retention strategy


There are two basic methods for sample treatment: a. Select the packing bed to retain the desired analyte. The contaminants are washed off and the desired analyte is then eluted for analysis. b. Select the packing bed to retain the contaminants and the desired analyte passes directly through.

Process multiple samples on a vacuum manifold using SPE columns. Extend volume via reservoirs.

3. Select the proper packing type


Select the proper packing type for the cleanest extract with the highest recovery. a. Reversed-phase packings are hydrophobic, silica-based materials that retain moderately polar to non-polar compounds from a polar matrix while washing off polar interferences. Or you can retain non-polar contaminants while the polar compounds pass through unretained. b. Normal-phase packings are hydrophilic, silica-based materials that retain polar compounds from a nonpolar matrix while washing off non-polar interferences. Or you can retain polar contaminants while non-polar compounds pass through unretained. c. Ion-exchange resins retain charged compounds or remove ionic interferences.

4. Optimize conditions for best results


Select proper bed size and suitable conditioning, wash and elution solvents. a. Poor sample recovery often occurs when the packed bed dimensions are not optimized. An excessive bed weight results in incomplete elution while an insuf cient bed weight results in incomplete retention. b. Consider the solvent strength relative to the packing material. The conditioning solvent should be nal weak so it doesnt act as an eluting solvent. Buffers should be used to control ionization of potentially charged compounds. c. Wash solvents should remove weakly retained interferences without being strong enough to elute the analyte. d. Elution solvents should be strong enough to completely elute an analyte in a small volume (12mL).

The resin bed mass determines the maximum amount of analyte retained by the sorbent. Tips: Choosing the proper bed mass reduces leaching of sample components

Tips for Choosing Proper Resin Mass Polymeric Sorbentspolymeric resins provide larger surface area than silica-based resins, offering higher capacity on the order of 10-15% of the resin mass. This allows for the use of smaller bed volumes and improved detection limts. Ion Exchange Sorbentsthe number of charged sites on the resin available to interact with the analyte determines capacity.
Tip: Choosing a slightly larger resin mass is advisable, since most SPE samples are loaded with contaminants and interferences

Resin Bed VolumeInterstitial and Pore Volume The resin bed volume controls the minimum solvent volume in an SPE method. To successfully condition, wash and elute from the cartridge typically requires solvent volumes 4 to 8 times that of the sorbent bed. Using less than 4 to 8 sorbent bed volumes contributes to incomplete solvation and low, inconsistent recoveries.
Tips: Polymeric SPE products usually require 200-250 uL per 100 mg of sorbent.

Universal Robotic Manifold

Based on the successful MicroLute Acrylic Manifold, the new Universal Robotic Manifold is designed to be easily assembled and disassembled by robotic manipulators, enabling productive automation of SPE or DNA clean-up procedures. Able to accommodate collection plates from 14mm 44mm in height and adaptable for working with short, medium and long skirts and drip directors, the Universal Robotic Manifold can operate with many different brands of filter plate and collection trays. Supplied with an integral vacuum gauge, the Manifold provides complete control of vacuum pressure ensuring plates are processed with high reproducibility.

The Manifold is fully compatible with most commercial robotic liquid handling systems. The Universal Robotic Manifold is suitable for a range of manufacturers plates without an adaptor. Vacuum Manifolds
Vacuum manifolds for SPE sample preparation, filtration, and elusion are available in 12, 16, and 24 port configurations from Ciro Manufacturing Corp. These manifolds permit consistent extraction and filtration results. Multiple sample processing with these manifolds simplifies procedures and saves time.The manifolds consist of a clear glass chamber and lid to which a vacuum is applied to draw a sample through an SPE column, cartridge, or disk. Adjustable racks placed in the glass vacuum chamber will accommodate a variety of sample collection vessels, including test tubes, autosampler vials, volumetric flasks, and Erlenmeyer flasks. Eluants are deposited directly into the collection vessel of choice via polypropylene, optional stainless steel, or Teflon needles. Drying attachments for the 12 and 24 port manifolds will direct a flow of air or nitrogen into the collection vessels to dry eluants prior to further analysis. Drying attachments can also be connected, via adapters, to SPE columns or cartridges in order to dry the column or cartridge prior to final elusion. Optional disposable solvent resistant polypropylene liners are available for the twelve port manifolds.These waste liners greatly simplify sample preparation, solvent disposal, and clean-up. Drying Attachments are available for the 12 and 24 port manifolds, which will direct the flow of air or nitrogen into the collection vessels to concentrate eluants, prior to further analysis. Drying attachments can also be connected directly to columns, cartridges, or disks, via adapters to permit drying sorbent beds, prior to final elution. Disposable Teflon needles and Teflon needles with flow control valves are designed to fit through the manifold lid via the luer fitting.These needles deliver the eluant directly from the SPE column or cartridge into the collection vessel in the vacuum chamber.These needles, when used in conjunction with Teflon columns and Teflon frits ensure zero extractables from the column, frits, and fluid path. This combination is especially useful for critical sample analysis, such as environmental samples. Disposable solvent resistant polypropylene waste liners, for 12 port manifolds, greatly simplify sample preparation, solvent disposal, and final clean-up of the manifold. The disposable waste liner is a molded solvent resistant polypropylene liner that fits into the vacuum chamber of the 12 port manifold. The liner is designed to contain all liquids used in SPE sample preparation.To use the liner, remove the manifold lid and take out the rack and shelf set. Place the disposable liner into the glass vacuum chamber, and replace the manifold lid. Proceed with all conditioning and sample preparation steps. Just prior to final elution, the liner, containing the waste solvents, is removed from the vacuum chamber. There are small handles at each end of the waste liner to facilitate its removal. Replace the lid, and proceed with the final elution of SPE cartridges are available in a wide variety of the analyte. Waste solvents should be properly discarded from the liner. The liner can be cleaned and re-used a number of times, prior products. to discarding.

Porvair evaporators are designed to remove the traditional laboratory bottleneck of solvent evaporation from microplates prior to analysis or reconstitution in storage buffer. These evaporators give significant throughput advantages to laboratories looking to optimize microplate sample preparation productivity. Faster than centrifugal evaporation, significant increases in sample throughput are achieved through advanced evaporator head technology and an innovative manifold design, which directly injects heated nitrogen into each individual well of the microplate simultaneously. EVX - 96 Evaporex

The Evaporex series solvent evaporators offer high-performance drydown for microplates at a reasonable price. The Evaporex has two needle assemblies, each consists of 96 individual stainless steel needles that evenly distribute heated gas into the microplates from both the top and bottom. The upper gas flow path is sealed away from the heating elements in a stainless steel path in order to prevent sample contamination. The lower platform can be adjusted to any height from the upper needle assembly, allowing the needles to come in close proximity to - without actually touching - the sample. This feature minimizes drying time and enables the user to develop methods utilizing a variety of plate sizes and sample volumes. The EVX Series is suitable for use with aqueous or organic solvents (not dry or concentrated acid solutions).

Biological samples commonly contain proteins that interfe re with downstream applications. The P3 plate uses the CRAS H method, in which the protein is denatured with acetronitr ile and the flocculant filtered out, allowing 96 samples to be handled at one time. The Protein Precipitation Plate, P3, is based on the industry standard MicroLute format, but wi thout the chromatographic sorbent. Protein crashes out of solution and precipitates directly in each well when acetonitr ile is added, thus solving all common problems associated w ith the CRASH technique of protein clean-up. The novel dual f rit, hydrophobically treated matrix means that there is no wetti ng out and leakage of the sample through the plate before t he application of vacuum.

Syringe Filters
Filtration is achieved by pushing the sample through the membrane with a syringe or other luer-connection device. Syringe lters allow you to control the rate of ow, which can be critical with delicate samples. Syringe lters also allow you to lter into nearly any tube, vial, or column that represents the next step in your analysis.

Centrifuge Filter Tubes


Filtration is achieved by pulling the sample through the membrane with centrifugal force. This allows you to simultaneously lter as many samples as your centrifuge can hold, and captures your ltrate in a clean microtube.

Pre-Cut Membrane

These convenient, high performance filter holders consist of a borosilicate glass funnel (300mL graduated) and base (with a fused-in coarse porosity fritted support; anodized aluminum clamp and a #8 silicone stopper to fit 1000mL filter flasks). Mating surfaces are precisely ground flat to assure a leak-resistant seal. The glass parts and the clamp are designed to work together to prevent any wrinkling or tearing of the filter membrane. Why are 40 mg/L PAR comparisons important?


EPA Method 1664A requires the PAR concentration as follows: The weight must be 40 1 mg (Section 7.10.3 NOTE) and at least this amount are required for the QCS, OPR, MS, MSD, and IPR samples. (Sections 9.8, 9.7, 9.6.1, 9.3.1, 9.2.2, 7.11 and 7.10) Although certain method modifications are allowed, a change in the PAR concentration is not expressly permitted and shall be considered a major modification subject to application and approval of alternate test procedures under CFR 136.4 and 136.5.
