Research Biochemist &Research Associate/Department of Diabetes, Rahway, NJ (1999 2007) Managed, developed, optimized and provided enzymology assay support for the development of potential Type II diabetes and obesity small molecule drug candidates. Generated, analyzed, reported, and coordinated data cross-departmentally with animal research and medicinal chemistry. Developed, optimized, implemented and supported an assay for adipose derived protein Acrp30 for use as a potential phase I clinical biomarker for the identification of Type II diabetes. o Determined the therapeutic effectiveness of small molecules approximately 10 times faster than animal studies by correlating plasma/serum Acrp30 levels to glucose lowering. Awarded Merck Rahway Research Recognition Award in 2005-2006. Isolated RNA for molecular profiling for the identification of new small molecule targets in Diabetes and Obesity. Identified and characterized a 10-fold more sensitive substrate for the Insulin Receptor Activator program, increasing the assay sensitivity and window of measurement for small molecule screening. Awarded Merck Special Achievement Award in Diabetes in 2000. Developed both a radioactive TCA precipitation and HPLC assays for the Insulin Receptor Activator program to quantify bound vs. unbound substrate. Contributed to the set-up and development of the Murein pathway assay of the 6 Stage I enzymes involved in bacterial cell wall biosynthesis, which increased the screening throughput of small molecules 6-fold. o Featured in the American Chemical Society January 4, 1999 publication Chemical & Engineering News. o Automated and miniaturized screening for the discovery of new and novel antibiotics. o Received the Merck Award of Excellence in 1996 and a patent for this methodology. o Developed HPLC methods for each individual Mur enzyme (C-F) for fast and easy target identification. Assayed and titrated small molecule compounds towards the development of a selective inhibitor of inducible Nitric Oxide Synthase (iNOS) for the treatment of rheumatoid arthritis. o Investigated the mechanism of action of iNOS using kinetic and spectroscopic methods.
