A group of student carried out a experiment to show the effects of varying the temperature on the rate of
an enzyme-catalysed reaction. They used amylase as the enzyme and starch as the substrate. Mixtures of
amylase and starch suspensions were immersed in water baths set at 5 oC as shown in diagram 1.1
At every two minutes interval, a drop of the mixture was transferred onto a tile containing iodine solution.
The step was repeated for mixtures at 28 oC, 37 oC, 45 oC, and 60 oC. Table 1 shows the colour of the
iodine solution in the tiles at different temperatures.

Microorganisms are very small organisms that can only be seen clearly under a microscope. The activity of
microorganisms is affected by abiotic components such as concentration of nutrients, light intensity, pH and
temperature of the surroundings.

Design a laboratory experiment to study the effect of temperature on the activity of yeast. The planning of
the experiment must include the following aspect :
-

Statement of identified problem

Objective
Statement of hypothesis
Variables
List of apparatus and materials
Technique used
Experimental procedure
Presentation of data
Conclusion
