GLUTATHIONE AS ANTIOXIDANT

IN PREVENTION OF AGING

WHAT IS GLUTHATHIONE?
Glutathione (GSH) is often referred to as the body's master
antioxidant.

Composted of three amino acids - cysteine, glycine, and


glutamate - glutathione can be found in virtually every cell of
the human body.
The highest concentration of glutathione is in the liver, making
it critical in the body's detoxification process.

Glutathione has multiple functions


The major functions of glutathione in the body include
protecting cells against the destructive effects of free radicals
detoxifying external substances such as drugs, environmental
pollutants and carcinogens
maintaining cell membrane stability
regulating protein and DNA biosynthesis and cell growth
enhancing immunologic function through its influence on
lymphocytes
prostaglandin synthesis; and amino acid transport

BIOCHEMISTRY OF GLUTHATHIONE
Glutathione exists in both reduced (GSH) and oxidized (GSSG) states.
Reduced Glutathione (GSH) is a linear tripeptide of L-glutamine, Lcysteine, and glycine. L-Glutathione, the molecule has a sulfhydryl (SH)
group on the cysteinyl portion, which accounts for its strong electrondonating character.
As electrons are lost, the molecule becomes oxidized, and two such
molecules become linked (dimerized) by a disulfide bridge to form
Glutathione disulfide or oxidized Glutathione (GSSG). This linkage is
reversible upon re-reduction.
Mitochondrial glutathione is critical to the
healthy cell, and is probably the most
important antioxidant defense system within
the mitochondria. Age-related alterations in
these enzymes can have a profound adverse
effect on health

GLUTATHIONE AS ANTIOXIDANT

An antioxidant is a molecule that inhibits the oxidation of other molecules. Oxidation is a


chemical reaction that transfers electrons or hydrogen from a substance to an oxidizing agent.
Oxidation reactions can produce free radicals. In turn, these radicals can start chain reactions.
When the chain reaction occurs in a cell, it can cause damage or death to the cell.

Antioxidants terminate these chain reactions by removing free radical intermediates, and inhibit
other oxidation reactions. They do this by being oxidized themselves, so antioxidants are often
reducing agents such as thiols, ascorbic acid, or polyphenols

Glutathione has antioxidant properties since the thiol group in its cysteine moiety is a reducing
agent and can be reversibly oxidized and reduced.

In cells, glutathione is maintained in the reduced form by the enzyme glutathione reductase and
in turn reduces other metabolites and enzyme systems, such as ascorbate in the glutathioneascorbate cycle, glutathione peroxidases and glutaredoxins, as well as reacting directly with
oxidants.

Due to its high concentration and its central role in maintaining the cell's redox state, glutathione
is one of the most important cellular antioxidants.
